The polymorphism of HLA‐A, ‐C, ‐B, ‐DRB3/4/5, ‐DRB1, ‐DQB1 loci in Zhejiang Han population, China using NGS technology

Abstract: The distributions of HLA allele and haplotype are various in the populations. Currently, the data for HLA alleles and haplotypes at three fields resolution level in Chinese Han population is rare. Here, the HLA alleles and haplotypes of the 1734 cord blood samples from Zhejiang Han population, China were reported at three fields resolution. All samples were randomly collected from the Zhejiang Cord Blood Bank, China. HLA-A, -B, -C, -DRB1, -DQB1, -DRB3/4/5 loci was genotyped using next generation sequencing met… Show more

“…The distribution of HLA-A , HLA-B , and HPA systems was similar to those of our previous reports ( 11 , 35 ). Furthermore, some SNVs were detected in the coding region, which is located in the structural domain of the platelet membrane glycoproteins ( Table 3 ).…”

“…At present, many DNA-based methods have been used for HLA and/or HPA genotyping in platelet donors ( 20 28 , 30 , 31 ). It has been reported that PCR-SBT and NGS methods have been routinely used for HLA genotyping in the laboratory ( 24 26 , 35 , 36 ). The NGS methods for HLA genotyping are divided into targeted and amplicon-based methods ( 34 , 35 ).…”

“…The HLA‐A, ‐C, ‐B, ‐DRB1 and ‐DQB1 loci of the donor were genotyped using the AllType™ NGS 9‐loci amplification kit (One Lambda Inc., Canoga Park, CA, USA) and the genotype of the donor was assigned using a TypeStream Visual Software version 1.2.0(One Lambda Inc.) as per our previous reports 2–4 . According to the sequence result of NGS, a novel HLA‐B allele, now officially named as HLA‐B*46:95N was identified.…”
