The organization of Physcomitrella patens RAD51 genes is unique among eukaryotic organisms

Abstract: Genetic recombination pathways and genes are well studied, but relatively little is known in plants, especially in lower plants. To study the recombination apparatus of a lower land plant, a recombination gene well characterized particularly in yeast, mouse, and man, the RAD51 gene, was isolated from the moss Physcomitrella patens and characterized. Two highly homologous RAD51 genes were found to be present. Duplicated RAD51 genes have been found thus far exclusively in eukaryotes with duplicated genomes. Ther… Show more

“…For DNA gel blot analysis, nuclei prepared from Col-0, fas1-43N1IC4 651, and N1IC4 651 plants were mixed before DNase I digestion as indicated and digested as described above. After extraction, the DNA was digested with EcoRI, and fragments were separated by agarose gel electrophoresis, blotted onto a nylon membrane, and hybridized with radioactively labeled probes as described (Markmann-Mulisch et al, 2002).…”

“…The P. patens genome contains two duplicated, functional RAD51 genes, Pp RAD51A and Pp RAD51B (Markmann-Mulisch et al, 2002), also described as PpaRad51.1 and PpaRad51.2, both of which encode genuine RAD51 proteins with all attributes of RAD51 (Ayora et al, 2002). To analyze the role of RAD51 in development and DNA damage repair in this plant, we used gene targeting to knock out both RAD51 genes.…”

“…Bruch & Schimp has been established as an important model species of primitive moss plants. Major progress has been made in recent years in analysing the molecular biology and physiology of this species (eg Markmann-Mulisch et al, 2002;Bezanilla et al, 2003). A substantial breakthrough was achieved with the demonstration that homologous recombination occurs in its nuclear DNA at frequencies which are orders of magnitude greater than those known from higher plants (Schaefer and Zrÿ d, 1997).…”
