The NR4A family of orphan nuclear receptors are not required for adipogenesis

Abstract: Nuclear hormone receptors of the NR4A subfamily are rapidly induced during the early stages of adipogenesis, leading to the speculation that they may have important roles in this process. One of the three subfamily members, Nur77 has also been shown to play key roles in energy expenditure and lipolysis in skeletal muscle and in the control of hepatic gluconeogenesis. We, therefore, examined the role of NR4A factors in adipogenesis using the well-characterized 3T3-L1 preadipocyte model. Inhibition of Nur77 expr… Show more

“…As an additional control, we also tested the effect of Nur77-898, an N-terminally truncated dominant-negative receptor (deletion of the first 897 nucleotides of Nur77). Deletion of the N-terminal transactivation domain has previously been shown to antagonize the activity of all three NR4A isotypes (12, 17, 28). Following puromycin selection of large mixed pool of cells (not single clones), each cell line was differentiated in standard differentiation cocktail for 7 days (DMI for 2 days, followed by insulin alone for 5 days).…”

“…Others have reported, however, that induction of NR4A receptors occurs some 24 to 48 hours after the addition of the PPARγ ligands troglitazone or pioglitazone to fully differentiated adipocytes (16). On the other hand, Au et al have suggested that NR4A receptors are not required for adipogenesis (17). Collectively, these studies raise the possibility the NR4As may be involved in the regulation of adipocyte gene expression during differentiation.…”

“…Initial studies using siRNA approaches and overexpression of a Nur77 mutant lacking the N-terminal AF-1 transactivation domain indicated that Nur77 is not required for adipocyte differentiation 55. However, a functional role for NR4A receptors in adipogenesis was suggested by two recent in vitro studies, which have demonstrated that constitutive NR4A receptor expression in 3T3-L1 preadipocytes inhibits adipocyte differentiation 56, 57.…”
