The New Era of the Lymphatic System: No Longer Secondary to the Blood Vascular System

Abstract: The blood and lymphatic systems are the two major circulatory systems in our body. Although the blood system has been studied extensively, the lymphatic system has received much less scientific and medical attention because of its elusive morphology and mysterious pathophysiology. However, a series of landmark discoveries made in the past decade has begun to change the previous misconception of the lymphatic system to be secondary to the more essential blood vascular system. In this article, we review the curr… Show more

“…Identification of the same TSC2 mutation in LAM and AML in patients bearing both lesions (Carsillo et al 2000) also supports a neural crest origin of these neoplasms. The media (smooth muscular layer) portion of the anterior cardinal vein was found to be neural crest derived (Bergwerff et al 1998;Adams and Alitalo 2007), and it is from this vein that PROX1-positive endothelial cells bud off to form rudimentary lymphatic vessels during embryonic development (Choi et al 2012). However, no studies have been conducted showing derivation of the endothelium of blood or lymphatic vessels from neural crest cells.…”

“…Although the hemovascular system has been studied extensively, lymphatics have been neglected. 1 "Lymph" is Latin for "connected to water," reflecting lymph's translucency, yet our understanding of lymphatics is unclear. Several recent studies have helped to clarify lymphatics' roles in health and disease.…”

“…Both blood and lymphatic vessels are lined with endothelial cells (ECs), which are critical for the function and maintenance of both vascular networks (1). Normal lymphatic vascular function is required for maintaining fluid balance, immune surveillance, and lipid homeostasis (2,3).…”
