The cultivation of white rot edible fungus Pleurotus florida was performed in polybags. The corn cob was employed as basal substrate while eight different additives such as urea, ammonium sulphate, gram flour, soybean meal, ground nut cake and molasses were used with corn cob. Three different levels of variable combinations with corn cob were evaluated in response to different parameters of mushroom viz., mycelial growth, spawn running, primordial initiation, fruit body yield and its biological efficiency. Each additive at different combinations showed variable impact on the different stages of mushroom life cycle. The primordial initiation was observed for the first time during 20.2-35.1 days. The biological efficiencies in every supplemented set were increased over un-supplemented control set. Increasing the level of additives, the biological efficiency was negatively affected at higher levels. The cotton seed cake was found the best supplement producing 93.75% biological efficiency while soybean meal was the second best additive producing 93.00% yield. The highest growth rate, rapid mycelia run, early primordial initiation, highest yield and biological efficiency were recorded in the combination of corn cob and cotton seed cake at 2% (98 + 2) level.

Analysis of 49 strains of F. langsethiae originating from northern Europe (Russia, Finland, Sweden, UK, Norway, and Latvia) revealed the presence of spontaneous auxotrophic mutants that reflect natural intraspecific diversity. Our investigations detected that 49.0% of F. langsethiae strains were auxotrophic mutants for biotin, and 8.2% of the strains required thiamine as a growth factor. They failed to grow on vitamin-free media. For both prototrophic and auxotrophic strains, no growth defect was observed in rich organic media. Without essential vitamins, a significant reduction in the growth of the auxotrophic strains results in a decrease of the formation of T-2 toxin and diacetoxyscirpenol. In addition, all analysed F. langsethiae strains were distinguished into two subgroups based on PCR product sizes. According to our results, 26 and 23 strains of F. langsethiae belong to subgroups I and II respectively. We determined that the deletion in the IGS region of the rDNA of F. langsethiae belonging to subgroup II is linked with temperature sensitivity and causes a decrease in strain growth at 30 °C. Four thiamine auxotrophic strains were found in subgroup I, while 21 biotin auxotrophic strains were detected in subgroups II. To the best of our knowledge, the spontaneous mutations in F. langsethiae observed in the present work have not been previously reported.

Termites form one-piece nests in wood that can vary in their moisture content and degree of decomposition, and thus microbial richness. To estimate the microbial load of nests and the potential risk they pose for colony members, we quantified the number of microbes in the nest and on the cuticle of the dampwood termite, Zootermopsis angusticollis , and three drywood termites, Incisitermes minor , I. schwarzi , and Cryptotermes cavifrons . The number of colony forming units (CFUs) cultured from nest material samples and washes of the cuticle of larvae and nymphs were determined. CFUs recorded from nest material was low (fewer than 60 CFUs/g) in the drywood termites and comparatively high in the dampwood species, as more than 800 bacterial and fungal CFUs/g were cultured from the nest material of Z. angusticollis . Similarly, cuticular microbial loads were negligible in the drywood termites sampled, ranging from 0.5 to fewer than 16 CFUs/cm 2 , whereas approximately 200 CFUs/cm 2 were cultured from Z. angusticollis . The nesting and feeding habits of these basal species likely influence colony microbial load and the degree of pathogen exposure, which in turn could favor adaptations to resist disease that vary with termite nesting biology.
