Identification and Characterization of Spontaneous Auxotrophic Mutants in Fusarium langsethiae

Abstract: Analysis of 49 strains of F. langsethiae originating from northern Europe (Russia, Finland, Sweden, UK, Norway, and Latvia) revealed the presence of spontaneous auxotrophic mutants that reflect natural intraspecific diversity. Our investigations detected that 49.0% of F. langsethiae strains were auxotrophic mutants for biotin, and 8.2% of the strains required thiamine as a growth factor. They failed to grow on vitamin-free media. For both prototrophic and auxotrophic strains, no growth defect was observed in r… Show more

“…P granules are localized adjacent to nuclear pores, to survey the expression of mRNA molecules exiting the nucleus 22,31,32 . Various P granules-associated proteins were found to be required for RNAi or for RNAi inheritance, including the P granule core-forming proteins PGL-1 33,34 , MEG-3 and MEG-4 20 , and the VASA homolog GLH-1 35 . Several argonaute proteins are also localized to P granules including PRG-1 11 , WAGO-1 19 , CSR-1 17 .…”

“…Previously, histone-H3-lysine-9 (H3K9) methylations were implicated in trans-generational inheritance of small RNAs [47][48][49][50]. H3K9 mono-and di-methylations are deposited by the MET-2 methyltransferase [51], while H3K9me3 is deposited by the SET-25 and SET-32 methyltransferases [51,52].…”
