The MYST histone acetyltransferases are essential for gametophyte development in Arabidopsis

Abstract: Background: Histone acetyltransferases (HATs) play critical roles in the regulation of chromatin structure and gene expression. Arabidopsis genome contains 12 HAT genes, but the biological functions of many of them are still unknown. In this work, we studied the evolutionary relationship and cellular functions of the two Arabidopsis HAT genes homologous to the MYST family members.

“…S6). Also, HAM1 and HAM2 are functionally redundant, as single Arabidopsis ham1 and ham2 mutants display a wild-type phenotype, while no double mutant seedling can be recovered (Latrasse et al, 2008). In our experiments, after 4 h of UV-B, both genes are induced in wild-type plants (Fig.…”

“…S2), showing that the HAM1 transcript abundance in ham1-2 was 2-fold lower than in wild-type plants. For the HAM2 gene, a T-DNA insertion in the second intron was obtained from the Delarue laboratory (Latrasse et al, 2008; SALK_106046; ham2-1). Supplemental Figure S2 shows that homozygous plants had undetectable levels of HAM2 transcripts when analyzed by RT-PCR.…”

“…AtHDA7 is required specifically for female (not for male) gametophyte development in Arabidopsis, particularly at the four-nucleate stage. Only HAM1 and HAM2, belonging to MYST HATs, were described as essential for female as well as male gametophyte development in Arabidopsis (Latrasse et al, 2008). In HAM1/ham1; ham2/ ham2 and ham1/ham1; HAM2/ham2 plants, half of the ovules aborted, since megagametogenesis failed to progress beyond the one-nucleate stage.…”

“…Similarly, the transcription factors CO, MRG1/ MRG2, and HAM1/HAM2 likely also form a module in promoting FT transcription and plant flowering Xu et al, 2014), albeit it is remaining to be verified whether CO is required for H4K5ac deposition. Because the ham1ham2 mutant is lethal (Latrasse et al, 2008), direct examination of function of H4K5ac deposition by HAM1/HAM2 in either the PIF7-MRG1/ MRG2 or CO-MRG1/MRG2 module has not been performed so far. Differing from H4K5ac, H3K9ac responded to shade induction in only a PIF7-dependent but not a MRG1/MRG2-dependent manner.…”
