The Mink Circovirus Capsid Subunit Expressed by Recombinant Baculovirus Protects Minks against Refractory Diarrhea in Field

Abstract: Mink refractory diarrhea is a seasonal disease that occurs in many mink farms in China. Mink circovirus (MiCV) has been recognized as the causative agent of the disease. The aim of the study was to develop a subunit vaccine against mink refractory diarrhea. A recombinant baculovirus strain expressing the capsid protein was constructed using the baculovirus expression vector system (BEVS). A subunit vaccine was developed based on the capsid protein with appropriate adjuvant. Then, a field trial was carried out … Show more

“…The results showed that the sensitivity of qPCR was higher than that of conventional PCR to detect MiCV. Moreover, the minimum detection limit of the qPCR method established in this study was lower than that of the previously established conventional PCR method and qPCR method ( Wang et al, 2021 ). The values of the intra-assay CV ranged from 0.46 to 0.96%, and the values of the inter-assay CV ranged from 1.37 to 1.98%, which indicated that the qPCR method is highly reproducible.…”

“…Some farmers used homemade inactivated vaccines (tissue supernatants of diseased mink inactivated using formalin) to prevent the disease and reduce morbidity; however, after a period of application, some minks developed symptoms of Aleutian disease. There are serious safety concerns with this type of vaccine; therefore, it has been discontinued ( Wang et al, 2021 ). In view of this situation, the effective way to prevent the disease is to detect and eliminate the positive animals gradually in China.…”

“…Some authors have reported [ 16 ] that in the case of influenza vaccines, employing BEVS instead of the classic embryonated egg platform, the time needed to produce 425 million vials of the vaccine would be 45 days instead of the average six months. FluBlok® [ 17 ] (Protein Sciences Corporation) is a quadrivalent influenza vaccine produced in BEVS by the expression of the hemagglutinin (HA) proteins of four different strains of influenza A and B viruses and is currently FDA-licensed for individuals 18 years old and older.…”

“…The subunit vaccinations have several advantages, including good specificity, reduced side effects, and greater safety. In order to maximize the effectiveness of most subunit vaccinations, immunopotentiators must be added, such as influenza vaccines ( Khalaj-Hedayati et al, 2020 ), COVID-19 vaccines ( Mekonnen et al, 2022 ), and mink virus vaccines ( Wang et al, 2021 ), etc.…”
