The Metabolism of Ring-Labeled L-Thyroxine-C14in Vivo

Abstract: In the past, investigators have used thyroxine labeled either with JIP" in the 3',5'-position or with C14 in the carboxyl position to study the metabolism of thyroxine in vivo. Although these studies have yielded much valuable information, our knowledge about the metabolic fate of the ring structure in thyroxine is incomplete because both the 3'5'-IJ13 and the carboxyl-C14 are rapidly removed from thyroxine in vivo by deiodination and .decarboxylation. To investigate the metabolic fate of the thyroxine nucleus… Show more

“…These findings suggest that thyronine and metabolite A2 had similar DISCUSSION After the administration of "C-labeled thyroxine in subreplacement amounts, the cumulative recovery of '4C in the urine was approximately 41% of the total given dose 2 wk after the last intravenous injection. This recovery was comparable with that reported by West, Simons, Gortatowski, and Kumagai (12) and that reported by us in other studies (8). The '4C-labeled metabolites A, and A2 were purified by repeated chromatography.…”

“…West et al (13) also observed that in the dog the disappearance rate of "C was much faster than that of 1311. Perhaps there are species difference in the handling of inorganic iodide and the deiodinated thyroxine product(s).…”

“…WVest and colleagues (13) reported that after the administration of [0-"C1 T4 to man the "C recovery from expired air was extremely small. After the administration of tracer amounts of 1I-labeled thyroxine to man, the major constituent of the urinary radioactivity has been found to be inorganic iodide, although thyroxine as well as iodinated tyrosines have also been observed during some earlier investigations (19,20) when pharmacologic doses of radiothyroxine were employed.…”
