The aim of this study was to find the biochemical markers of blackleg, evoked by the fungus Leptosphaeria maculans in seedlings of oilseed winter rape. Analyses of antioxidant enzymes activity and reactive oxygen species detection were performed 24, 48 and 72 h after inoculation. Moreover, measurements of the total pool of phenolic compounds, transcinnamic acid and ferulic acid were carried out. The results proved the virulence of Ph L5 isolate towards the rape seedlings. An increase in total phenolic content, including trans-cinnamic acid, was observed 24 h after inoculation. High activity of phenylalanine ammonia-lyase is probably another symptom of the defense mechanisms, despite the fact that it was not accompanied by elevated phenolics level, which were probably rapidly exhausted during Phoma lingam inoculation. A drop in phenolics content was balanced by higher activity of SOD, CAT and non-specific peroxidases. Enhanced activity of these enzymes was a response to the reactive oxygen species overproduction. ROS accumulation in indirectly inoculated first true leaves after inoculation may suggest a systemic defense response.

Reactive oxygen species (ROS) have been studied for their role in plant development as well as in plant immunity. ROS were consistently observed to accumulate in the plant after the perception of pathogens and microbes and over the years, ROS were postulated to be an integral part of the defence response of the plant. In this article we will focus on recent findings about ROS involved in the interaction of plants with pathogenic fungi. We will describe the ways to detect ROS, their modes of action and their importance in relation to resistance to fungal pathogens. In addition we include some results from works focussing on the fungal interactor and from studies investigating roots during pathogen attack.

In the present work we aimed at comparing the effect of benzothiadiazole (BTH) treatment on defence reactions of sunflower plants to downy mildew and white rot diseases. BTH treatment resulted in reduced disease symptoms in biotrophic and in the early stage of the necrotrophic interactions. To get a better insight into the effect of BTH, changes in the activities of polyphenol oxidase and peroxidase enzymes, as well as the expression of the host response-associated sunflower genes were examined in the plants. Inoculation with Plasmopara halstedii enhanced the polyphenol oxidase and peroxidase enzyme activities, while inoculation with Sclerotinia sclerotiorum did it only at 4 dpi. However, most importantly, in each case extracts from BTH pretreated and inoculated plants showed the highest polyphenol oxidase and peroxidase enzyme activities. Similarly, the accumulation of GST and PDF transcripts was detected following inoculations with both biotrophic and necrotrophic pathogens, and again, BTH pre-treatment enhanced GST and defensine gene activities in the inoculated plants. We suggest that induction of enzyme activities, as well as of the elevated expression of GST, PDF and PR5 genes by BTH pre-treatment may be a significant part of the induced resistance of sunflower to downy mildew and white rot (white mold).
