There are some of the problems faced in Biotechnology Subject. This research was aims to describe the preliminary research about the development of a local wisdombased Biotechnology module. It has been carried out with a 4-D model, which is a preliminary research. It show that students assume that the material about nibung is needed in the Biotechnology Subject. They assume that material in Biotechnology was in difficult category (87%). Furthermore, the students rarely learning Biotechnology other than when they are in the class (74%). Overall, students only use learning resources from internet and generally only rely on teaching materials from lecturers (100%). Students assume that it is needed to develop the teaching materials that can help students in independent study, as well as teaching materials that discuss about local commodities. In conclusion, there is a need to develop teaching materials containing research results based on local wisdom for Biotechnology Subject.

Due to the rapid development of industry in the market economy, a series of pollution problems have been brought accordingly. Among them, industrial sewage, domestic sewage treatment and other problems have seriously damaged the ecological environment and become one of the most critical issues to be solved for environmental protection nowadays. Therefore, this paper adopts biotechnology to treat domestic wastewater and explores the removal rate of COD, ammonia nitrogen, TP and TN in wastewater by displaced biochemical tank wastewater treatment process, and compares the treatment effect of displaced ball packing and combined packing with ordinary combined packing as reference. The results showed that the removal rate of each pollutant increased with the increase of water retention time of the displaced biochemical tank and the increase of the amount of filler in the biochemical tank, and the removal effect of the displaced ball filler on pollutants was significantly better compared with the combined filler.

This study aims to obtain an overview of the formulation of student worksheed (called as LKPD) based on local wisdom that needs to be developed in learning the biology of conventional biotechnology materials at SMA N 5 Kupang City which is adjusted to the learning achievements and objectives. The subjects in this study were 3 teachers who taught in class X of SMAN 5 Kupang City, and 175 students were randomly drawn from all grade X students of science interest at SMA N 5 Kupang City. Research data were collected through interviews and questionnaire dissemination. The research data were analyzed in a qualitative descriptive manner. The results of the study obtained teacher and student responses including students need LKPD based on local wisdom on biotechnology material to make it easier to understand the material, In addition, in learning, teachers provide examples of the results of biotechnology based on local wisdom in the form of pictures and videos, and are presented in the form of virtual laboratories with animations to attract more students' attention, so that students are more motivated to learn which will affect learning outcomes. The biology material for class X semester that is most difficult for students to understand is biological technology innovation material, so teachers need LKPD based on local wisdom in Biotechnology material, which can help students introduce local wisdom in their respective regions so that students can easily apply it in everyday life, and to support learning, which has a positive impact on improving student learning outcomes
