The inhibitory role of recombinant P‑selectin glycoprotein ligand immunoglobulin G on portal vein thrombosis based on a novel rat model

Abstract: The current study aimed to reveal a novel method for constructing a portal vein thrombosis (PVT) model in rats and to evaluate the inhibitory role of recombinant P-selectin glycoprotein ligand immunoglobulin G (rPSGL-Ig) on the formation of PVT. The PVT model was constructed in rats through intermittent portal vein obstruction (IPVO) combined with endangium destruction. A total of 4 mg/kg rPSGL-Ig was intraperitoneally pre-injected into rats 1 h prior to model construction. Changes in the thrombus size and ves… Show more

“…Male SD rats, weighing about 200–250 g, were used in this experiment, purchased from the Laboratory Animal Center of Lanzhou University and approved by the Animal Care and Use Committee of the First Hospital of Lanzhou University. The rat PVT model and hucMSC intervention were performed with reference to previously described methods 21 , 22 . Finally, the rats were randomly divided into five groups: (1) sham-operated group, (2) PVT group, (3) PVT + hucMSCs group (hucMSCs group), (4) PVT + NC-hucMSCs group (NC-hucMSCs group), (5) PVT + miRNA-25-3p-hucMSCs group (miRNA-25-3p-hucMSCs group).…”

“…Zhang et al [ 12 ] constructed a model by intermittently ligating the distal and proximal portal veins of rats to block blood flow. On this basis, Yao [ 13 ] and Wei et al [ 14 ] added clamping of blood vessels. They initiated the coagulation system by enhancing vascular endothelial cell injury, which induced PVT formation.…”
