The Inhibitory Effect of (−)-Epigallocatechin-3-Gallate on Breast Cancer Progression via Reducing SCUBE2 Methylation and DNMT Activity

Abstract: Epigenetic modifications are important mechanisms responsible for cancer progression. Accumulating data suggest that (−)-epigallocatechin-3-gallate (EGCG), the most abundant catechin of green tea, may hamper carcinogenesis by targeting epigenetic alterations. We found that signal peptide-CUB (complement protein C1r/C1s, Uegf, and Bmp1)-EGF (epidermal growth factor) domain-containing protein 2 (SCUBE2), a tumor suppressor gene, was hypermethylated in breast tumors. However, it is unknown whether EGCG regulates … Show more

“…Theaflavin 1 was found to prevent lung tumorigenesis via induction of apoptosis, down-regulation of fatty acid synthase and COX-2 in cellular and animal models. Theaflavin 2 induced cell death by regulating BAX and p53 protein in the HeLa and WI38VA cervical cancer cell line [45,52,53]. On figure below (Figure 4), we present a comparison of polyphenol content in green and black tea, broken down into theaflavins and catechins.…”

“…Overexpression of SCUBE2 reversed epithelial mesenchymal transition (EMT) and associated cellular signaling in breast cancer. A recent study by Sheng et al reported that EGCG significantly inhibited DNMT activity and demethylated SCUBE2 promoter methylation [62]. This resulted in higher expression levels of SCUBE2 and reduced breast cancer progression.…”

“…Current studies suggest that EGCG regulates DNA methylation and chromatin remodeling and alters the expression levels of different genes that regulate breast cancer proliferation and metastasis [58][59][60]. Following in vitro and in vivo studies, it was postulated that EGCG inhibits breast cancer proliferation and migration by regulating different tumor suppressor genes via its action of inhibiting DNA methyltransferases (DNMT) and histone deacetylases (HDAC) enzymes [61][62][63][64][65][66].…”

“…The natural polyphenol epigallocatechin-3-gallate (EGCG), found in green tea, was shown to inhibit DNMT activity by interacting with the enzyme catalytic pocket, reactivating the expression of several TSGs by reversing the hypermethylation status [ 116 , 117 ]. EGCG also decreased cell viability, migration and invasion of breast cancer cells, and SCUBE2 methylation by decreasing DNMT expression and activity [ 118 ]. Genistein is an isoflavone present in soybeans which displays anti-cancer activity [ 24 ].…”
