Quercetin and rutin as potential agents antifungal against Cryptococcus spp

Amphotericin B is a fungicidal substance that is treatment of choice for most systemic fungal infections affecting as cryptococcosis the immunocompromised patients. However, severe side effects have limited the utility of this drug. The aim of this study was to evaluate the antifungal effect of the combination of amphotericin B with quercetin or rutin and as a protective of citotoxic effect. The antifungal activity to amphotericin B, quercetin and rutin alone and in combination was determined in Candida sp and Cryptococcus neoformans strains. Cytotoxicity test on erythrocytes was performed by spectrophotometric absorbance of hemoglobin. The amphotericin B MIC was reduced when used in combination with quercetin or rutin to C. neoformans ATCC strain and reduced when combined with rutin to a clinical isolate of C. neoformans. In addition, the combination of quercetin with amphotericin B may reduce the toxicity of amphotericin B to red blood cells. Our results suggest that quercetin and rutin are potential agents to combine with amphotericin B in order to reduce the amphotericin dose to lessen side effects and improve antifungal efficacy.Keywords: amphotericin B, quercetin, rutin, Cryptococcus neoformans. Quercetina e rutina: potenciais agentes para terapia antifúngica ResumoA anfotericina B é uma substância fungicida e é o tratamento de escolha para a maioria das infecções fúngicas sistêmicas que afetam os pacientes imunocomprometidos, como a criptococose. No entanto, as severas reações adversas têm limitado a utilização desta droga. O objetivo deste estudo foi avaliar o efeito antifúngico e o potencial efeito protetor de citotoxicidade da combinação de anfotericina B com quercetina ou rutina. A atividade antifúngica de anfotericina B, quercetina e rutina, isoladamente e em combinação foi determinada em cepas de Candida sp e Cryptococcus neoformans. O teste de citotoxicidade em eritrócitos foi realizado por espectrofotometria, através da determinação da absorbância da hemoglobina. A concentração inibitória mínima da anfotericina B foi reduzida quando utilizada em combinação com a quercetina e rutina em C. neoformans ATCC e reduzida quando combinados com rutina em um isolado clínico de C. neoformans. Além disso, a combinação de quercetina com anfotericina B pode reduzir a toxicidade da droga em eritrócitos. Os resultados sugerem que quercetina e rutina são potenciais agentes para combinação com anfotericina B, a fim de reduzir a dose de anfotericina, diminuindo os efeitos colaterais e melhorando sua eficácia antifúngica.Palavras-chave: anfotericina B, quercetina, rutina, Cryptococcus neoformans.

ABSTRACT:Vulvovaginal candidiasis (VVC) is a common fungal infection that affects healthy women of all ages. At least 75% of women will develop one or more infections once during their lifetime, with 6 to 9% of those individuals developing recurrent infections. In view of this context, this study sought to evaluate the antifungal potential of the isolated (R)-(+)-citronellal [(R)-(+)-CT] and associated to therapeutic agents of clinical importance. The enantiomer was solubilized in tween 80 and dimethylsulfoxide (DMSO). Posteriorly diluted in sterile distilled water up to the concentration of 2048µg/mL. The minimum inhibitory concentration (MIC) of the product was determined by microdilution in RPMI-1640 obtaining dilutions of 1024-4µg/mL. The minimum fungicidal concentration (MFC) was determined by the Sabouraud dextrose agar (SDA) depletion technique from aliquots of 1µL of the MIC, MIC × 2 and MIC × 4. The MIC and the MFC values of (R)-(+)-CT for 90% of the C. albicans strains were 16 and 32µg/mL respectively. In the susceptibility test, C. albicans presented a high resistance to fluconazole and to itraconazole, 12 (92.30%) of the strains. However, for ketoconazole and miconazole the resistance was of 4 (30.76%) and 3 (23.07%) of the strains respectively. In the combination testing of the (R)-(+)-CT with ketoconazole and miconazole, the resistance was completely reverted. For fluconazole and itraconazole, the resistance was reverted in 9 (75%) and 7 (58.33%) of the strains respectively. The (R)-(+)-CT presented fungicide activity with MFC of MIC × 2. When in combination with ketoconazole, fluconazole, itraconazole and miconazole increased the inhibition zones of these antifungal drugs, reducing the resistance against C. albicans.

Abstract:The essential oil of Melaleuca alternifolia is an herbal antifungal action and preventive scale pharmaceutical or cosmetic. The aim of this study was to evaluate the efficacy of Melaleuca alternifolia (tea tree oil-TTO) and its component (Terpinen-4-ol) on Candida albicans and Candida tropicalis in planktonic and biofilm form of dual species. MIC (Minimum Inhibitory Concentration) and MFC (Minimum Fungicidal Concentration) were determined of single species. Biofilms were quantified in CFU (colony forming unit per mL) and XTT {2,3-bis (2-methoxy-4-nitro-5-sulfophenyl)-5-[(phenylamino) carbonyl]-2H-tetrazolium hydroxide} cell viability assay in single and multiple species. MIC was TTO (0.25%) and Terpinen-4-ol (0.11%) for both species. The MFC for Candida albicans showed TTO (0.25%) and Terpinen-4-ol (0.11%) were capable of inhibiting the growth and for Candida tropicalis was 0.50% and 0.24%, respectively. The results on biofilm for Candida albicans in single species and multiple species showed TTO (0.5%) and Terpinen-4-ol (0.24%) were effective. For Candida tropicalis in single species, treatments with TTO (0.5%) and Terpinen-4-ol (0.47%) were effective. For multiple species, the treatments with TTO (0.5%) and Terpinen-4-ol (0.24%) were also effective. In the XTT test, for Candida albicans in single species: TTO (0.25%) and Terpinen-4-ol (0.24%). For Candida tropicalis, the results were TTO (0.5%) and Terpinen-4-ol (0.11%), while for multiple species were TTO (0.25%) and Terpinen-4-ol (0.11%). Based on these factors, Candida spp. are susceptible to the antimicrobial agents TTO and Terpinen-4-ol, suggesting that these compounds might be beneficial as therapeutic.
