The inhibition by dexamethasone and disodium cromoglycate of anaphylactic bronchoconstriction in the rat

Abstract: Summary1. A method for the measurement of anaphylactic bronchoconstriction in the rat is described. 2. Dexamethasone inhibited this response in a dose-related manner. 3. Disodium cromoglycate antagonized the response. A bell-shaped doseresponse curve was obtained with peak activity at 1 mg/kg i.v. 4. Meclofenamate was not active when given alone or in combination with dexamethasone. 5. Methysergide significantly inhibited the response. Mepyramine, atropine, propranolol and adrenalectomy did not significantly m… Show more

“…3). Antagonists of serotonin and SRS-A, such as methysergide or FPL 55712 partially inhibit bronchoconstriction induced in rats (2,5,16). In the present experi ment, a partial inhibition of the bronchoconstriction in rats occurred following treatment with cyproheptadine.…”

“…For 3CA, intramolecular H-bonds (H· · ·O interactions, yielding a 6-membered ring) are clearly favoured over the intermolecular ones, the opposite being true for 2CA which is suggested to occur as a favoured top-to-top dimer in the condensed phase. This is in accordance with the extremely high pK a value previously reported for the 3-carboxylic chromone (8.85 [35] and presently calculated as 8.70), in contrast to 2CA's pK a (2.55 [36], currently calculated as 3.02).…”

“…Actually, this is the only species expected to occur at room temperature since the next calculated conformers correspond to very high relative energies ( E = 28.5, 34.1 and 65.0 kJ mol −1 , Table 1). Furthermore, this preferred intramolecular (O 10 )H· · ·O 12 ( C) close contact justifies the abnormally high pK a value reported for 3CA -8.85 [35] as compared to 2.55 [36] for the 2-carboxylic analogue, for which no such interaction is possible. These pK a values were presently confirmed by DFT calculations: 8.59 and 2.04 were obtained for the most stable conformers of 3CA and 2CA, respectively.…”

“…This is in contrast to the 'bell-shaped' dose-inhibitory response curve produced against anaphylactic bronchoconstriction in actively sensitized rats (Church, Collier & James, 1972).…”
