Mediators of Passive Lung Anaphylaxis in the Rat

Abstract: 1 Passive lung anaphylaxis (PLA) was investigated in rats sensitized by the intravenous injection of high titre reaginic antiserum prepared in rats. 2 The effect of various pharmacological antagonists on anaphylactic bronchoconstriction in vivo were examined. An antihistamine (mepyramine), a kallikrein inactivator (aprotinin) or a prostaglandin synthesis inhibitor (aspirin) did not inhibit PLA, whereas an anti-5-hydroxytryptamine agent (methysergide) and an anti-slow reacting substance-A agent (FPL, 55712) si… Show more

“…3). Antagonists of serotonin and SRS-A, such as methysergide or FPL 55712 partially inhibit bronchoconstriction induced in rats (2,5,16). In the present experi ment, a partial inhibition of the bronchoconstriction in rats occurred following treatment with cyproheptadine.…”

“…Rat anaphylactic bronchoconstriction, which is due to the release and action of 5-hydroxytryptamine and possibly slow reacting substance of anaphylaxis (SRS-A) (Church et al, 1972;Farmer, Richards, Sheard & Woods, 1975) is inhibited only by a high dose of dexamethasone and after a long lag period, possibly because the steroid depresses the release of mediators. Dexamethasone does not reduce the bronchoconstrictor effects of exogenous 5-hydroxytryptamine or SRS-A (Church, 1975).…”

“…Anaphylaxis-producing homologous antibody (IgE) can be produced in the Sprague-Dawley rats but this strain maybe cannot elicit bronchial constriction because of the lack of histamine receptor in the trachea of this strain. When Wistar rats were passively immunized with Sprague-Dawley-produced antibody, bronchial constriction occurred (Farmer et al, 1975). The current result indicates that Sprague-Dawley rats can be a model to study cardiovascular anaphylaxis without the involvement of bronchial system.…”
