The inheritance and chromosomal localization of AFLP markers in a non-inbred potato offspring

Abstract: AFLP T M is a new technique to generate large numbers of molecular markers for genetic mapping. The method involves the selective amplification of a limited number of DNA restriction fragments out of complex plant genomic DNA digests using PCR. With six primer combinations 264 segregating AFLP amplification products were identified in a diploid backcross population from non-inbred potato parents. The identity of an AFLP marker was specified by the primer combination of the amplification product and its size es… Show more

“…A great advantage of the AFLP approach is that it allows simultaneous identification of a large number of amplification products (Van Eck et al 1995;Vos et al 1995). Compared with RFLP or other PCR-based marker systems, AFLP is fast, reliable, and costeffective.…”

“…The integrated map of the C 9 E population consisting of 192 dominantly scored AFLP markers, 28 SSR markers and 24 CAPS/SCAR markers calculated as done previously (van Eck et al 1995). The linkage groups were assigned to the 12 linkage groups representing the 12 chromosomes on the basis of the earlier version of this map comprising a total length of 996 cm.…”

“…In this study an expanded version of the linkage map of van Eck et al (1995) was used, with an offspring size expanded to 249 full sib descendants. All additional individuals were genotyped using 16 AFLP markers with 15 primer combinations E32/ M47, E32/M48, E32/M49, E32/M50, E32/M51, E32/ M54, E32/M59, E32/M61, E35/M47, E35/M48, E35/ M50, E38/M59, E39/M60, E45/M59 and E45/M60.…”

“…These characteristics make AFLP a powerful fingerprinting technique which can be used in identification, epidemiology and taxonomy (Folkerstma et al 1996;Huys et al 1996;Janssen et al 1996). In addition, the technique can be used to generate large numbers of molecular markers for linkage studies (Ballvora et al 1995;Becker et al 1995;van Eck et al 1995).…”
