This study compared the fatty acid composition of wild and cultured specimens of the sea cucumber Apostichopus japonicus. We extracted total lipids from the specimens and determined their fatty acid compositions through capillary gas chromatography, resulting in the identification of 53 fatty acids. We found that wild sea cucumbers were rich in palmitoleic (C16:1n-7) and eicosapentaenoic acid (C20:5n-3), whereas cultured specimens were rich in eicosenoic (C20:1n-9) and arachidonic acid (C20:4n-6). In both types of sea cucumbers, the highest percentage of polyunsaturated fatty acids (PUFA) consisted of polar lipids (PL), followed by total lipids (TL) and neutral lipids (NL). Cultured sea cucumbers contained a higher percentage of total lipids (TL) than wild sea cucumbers did, whereas there was no significant difference between the two groups in the percentages of neutral (NL) and polar lipids (PL).

The effects of fermented fecal solid diets on the growth of the sea cucumber Apostichopus japonicus was investigated in a recirculating aquaculture system (RAS) during a 90-day experiment at 16.5-17.5°C, 32±0.5 psu salinity, pH 7.9-8.2, and 5.2-7.1 mg/L dissolved oxygen. The aim was to determine the feasibility of RAS fecal solid waste as a renewable feed for A. japonicus. In this study, unfermented fecal solid (UF), fermented fecal solid (FF), and commercial feed (CF) diets were used. The results showed that the growth rate did not differ significantly (P>0.05) between A. japonicus fed FF and CF; however, the growth rates of A. japonicus fed FF and CF were ~50% higher than that of those fed UF. Thus, utilization of fecal solid by A. japonicus may overcome the problem regarding accumulation of RAS waste. Use of a fermentation process would improve utilization of fecal solid as a renewable food source for A. japonicus.

Effects of different algae in diet on growth and interleukin (IL)-10 production of juvenile sea cucumber Apostichopus japonicus

The experiment was conducted to investigate the effects of different algae in diet on growth, survival, and interleukin-10 productions of sea cucumber. At first, a 9-week feeding trail was conducted to evaluate the growth performance and survival of the sea cucumber fed one of the six experimental diets containing ST (Sargassum thunbergii), UL (Ulva lactuca), UP (Undaria pinnatifida), LJ (Laminaria japonica), SS (Schizochytrium sp.), and NO (Nannochloropsis oculata) in a recirculating aquaculture system. The result showed that survival was not significantly different among the dietary treatments, and the specific growth rate (SGR) of sea cucumber fed the UL diet (1.58% d ) was significantly higher than that of sea cucumber fed the other diets (P < 0.05), except for the LJ and NO diets. Secondly, interleukin (IL)-10 gene expression was determined where mice splenocytes were stimulated with 10 μg ml −1 of sea cucumber extracts for 2 h. The result showed that IL-10 gene expression levels were significantly increased in UL, LJ, and NO diets fed sea cucumber extracts compared to other experimental diets. The results suggest that dietary inclusion with Ulva lactuca, Laminaria japonica, and Nannochloropsis oculata algae may improve the growth of juvenile sea cucumber and could upregulate IL-10 gene expression in mice splenocytes. Such detailed information could be helpful in further development of more appropriate diets for sea cucumber culture.
