The inflammasome and lupus

Abstract: Purpose of review The role of innate immunity in systemic lupus erythematosus (SLE) has been a rapidly expanding area of research over the last decade. Included in this rubric is the concept that activation of the inflammasome, a molecular complex that activates caspase-1 and in turn, the cytokines IL-1β and IL-18, is important in lupus pathogenesis. This review will summarize recent discoveries exploring the role of the inflammasome machinery in SLE. Recent findings Immune complexes can activate the NLRP3 i… Show more

“…Thus, cholesterol accumulation in ABC DKO DCs leads to inflammasome activation, increased secretion of IL-1β and IL-18 and skewing of T-cells towards a T h 1 phenotype. Interestingly, IL-18 levels are markedly elevated in human SLE, and inflammasome activation contributes to lupus glomerulonephritis in mice (Kahlenberg and Kaplan, 2014; Kahlenberg et al, 2014; Tucci et al, 2008), suggesting a role for inflammasome activation in the autoimmune phenotype of DC-ABC DKO mice.…”

“…The role of components of the innate immune response in the pathogenesis of systemic autoimmune diseases, such as lupus, has been increasingly recognized [15,16]. Lupus is characterized by activation of plasmacytoid dendritic cells, a robust type-I interferon expression signature, and hyper-responsiveness of PBMCs to type-I interferon [17,18].…”

“…75,76 Whether or not inflammasomes exert some sort of influence upon SLE's photosensitivity, however, is still under investigation, given that UVB radiation is capable of stimulating the synthesis of IL-1β in keratinocytes by activating the inflammasome. 77 …”
