The HLA-Cw*06 allele and −1149 G/T polymorphism of extrapituitary promoter of PRL gene as a possible common genetic predisposing factors to psoriasis vulgaris and psoriatic arthritis in Czech population

Abstract: Psoriatic arthritis (PsA) affects approximately 30 % of patients suffering from psoriasis vulgaris (PsV), but the risk factors for its development have not been well elucidated yet. The HLA-Cw*06 allele was described as a predisposing factor to PsV. Prolactin is known as an immune response modulator, and its elevated levels present risk for PsV development. It is possible that these factors interact and together emphasize the predisposition to both diseases. We tested on an association of HLA-Cw alleles and fu… Show more

“…Regardless of the differences of HLA‐Cw6 frequency in patients with PsA, HLA‐Cw6 is associated with early age of onset in both PsA and PsV. HLA‐Cw6 genotype is reported to be associated with early age of onset in patients with PsA, characterized by positive family history, severe psoriasis, clinical enthesitis, polyarthritis, oligoarthritis, axial pattern on disease onset and mixed pattern during evolution . For the other types of PsA, HLA‐Cw6 status is not associated with dactylitis in patients with PsA .…”

“…The worldwide HLA‐Cw6 allele frequency varies greatly and can range from 14·1% to 59·1%, whereas the percentage of patients with psoriasis carrying HLA‐Cw6 ranges from 10·5% to 77·2% (Table ) . Such wide ranges may be attributed to small sample sizes, ethnic diversity and differences in inclusion criteria.…”

“…Gudjonsson et al demonstrated that heterozygotes have a relative risk of developing psoriasis of 8.9, whereas homozygotes have a relative risk of 23.1, with earlier disease onset being more common in this group [11]. Cibulova et al found HLA-Cw*0602 to be the most prominent HLA in Type I PsV, correlating well with an earlier disease onset and a family history of psoriasis [12]. This finding was echoed in siblings discordant for HLA-Cw*0602, where the positive HLA-Cw*0602 sibling had significantly early disease onset [13].…”
