The Mental Health Biomarker Project (2010–2016) explored variables for psychosis in schizophrenia and schizoaffective disorder. Blood samples from 67, highly characterized symptomatic cases and 67 gender and age matched control participants were analyzed for methyl tetrahydrofolate reductase (MTHFR) 677C → T gene variants and for vitamin B6, B12 and D, folate, unbound copper, zinc cofactors for enzymes in the methylation cycle, and related catecholamine pathways. Urine samples were analyzed for indole-catecholamines, their metabolites, and oxidative-stress marker, hydroxylpyrolline-2-one (HPL). Rating scales were Brief Psychiatric Rating Scale, Positive and Negative Syndrome Scale, Global Assessment of Function scale, Clinical Global Impression (CGI) score, and Social and Occupational Functioning Assessment Scale (SOFAS). Analysis used Spearman’s correlates, receiver operating characteristics and structural equation modeling (SEM). The correlative pattern of variables in the overall participant sample strongly implicated monoamine oxidase (MAO) enzyme inactivity so the significant role of MAO’s cofactor flavin adenine nucleotide and its precursor flavin adenine mononucleotide (FMN) within the biochemical pathways was investigated and confirmed as 71% on SEM of the total sample. Splitting the data sets for MTHFR 677C → T polymorphism variants coding for the MTHFR enzyme, discovered that biochemistry variables relating to the wild-type enzyme differed markedly in pattern from those coded by the homozygous variant and that the hereozygous-variant pattern resembled the wild-type-coded pattern. The MTHFR 677C → T-wild and -heterozygous gene variants have a pattern of depleted vitamin cofactors characteristic of flavin insufficiency with under-methylation and severe oxidative stress. The second homozygous MTHFR 677TT pattern related to elevated copper:zinc ratio and a vitamin pattern related to flavin sufficiency and risk of over-methylation. The two gene variants and their different biochemical phenotypes govern findings in relationship to case-identification, illness severity, duration of illness, and functional disability in schizophrenia and schizoaffective psychosis, and establish a basis for trials of gene-guided precision treatment for the management of psychosis.

Determination of histaminase (diamine oxidase) activity byo-dianisidine test: Interference of ceruloplasmin

Until now o-dianisidine was used as an indicator substance in a test system for the determination of diamine oxidase. More recently, however, this substance was also used to measure ceruloplasmin activity. A study of the test principles revealed that o-dianisidine was the one denominator for both enzymes. As it was found for diamine oxidase the indicator was oxidized via peroxidase mediated H2O2 cleavage. Ceruloplasmin, however, oxidized o-dianisidine directly with resulting free radical formation. An addition of histamine dihydrochloride or putrescine dihydrochloride to an incubation mixture, containing ceruloplasmin as enzyme and o-dianisidine or p-phenylene-diamine as substrates, produced an activation of the enzyme, being more than 10-fold in the presence of 1 X 10(-2) M putrescine at pH 7.0. It was assumed that an allosteric effect of the dihydrochloride component might be responsible for this activation. When the activity of purified diamine oxidase was determined by the o-dianisidine test and by the isotope assay, a very good correlation between both methods was found. But, in a mixture of diamine oxidase and ceruloplasmin, no differentiation between the two enzymic activities by the o-dianisidine test was possible. This observation demonstrated an interference of ceruloplasmin when the o-dianisidine method was used for the determination of diamine oxidase activity. To apply our findings also in vivo the amine oxidase activity increasing in guinea-pig plasma during inflammation, was determined by the o-dianisidine test and by specific methods for some amine oxidase. Despite an enhanced oxidation of the o-dianisidine observed, only an increase of ceruloplasmin activity was found. It was concluded that ceruloplasmin had no 'histaminase activity' as has been assumed by other authors using the o-dianisidine test.

The plasma histaminase activity in guinea-pigs was found to increase by experimental inflammation induced by a subcutaneous turpentine injection and carrageenan peritonitis. The histaminase activity was raised 24 hours after the injection and did not return to normal values before 2 weeks. It was also observed that the alteration in the erythrocytes sedimentation rate followed a similar time course as that of the plasma level of histaminase. No relationship existed between the increased histaminase activity and skin oedema or changes in body temperature. The kinetics of the increase of plasma histaminase activity during inflammation are discussed.Since the classical experiments of LEwis [1] concerning the 'H substance', histamine has been commonly regarded as the basic mediator of inflammation. Histamine is possibly responsible for the initial as well as for the prolonged phase of increased permeability of the blood vessels in acute inflammation [2,3].In the first period of inflammation, histamine is liberated from the mast cells [4], whereas during the next period, an increased production of amine [5][6][7] takes place.The influence of inflammation on the behaviour of the enzymatic system catalizing the degradation of histamine has not as yet been thoroughly examined.In our previous experiments concerning the histaminase activity of plasma in guinea-pigs adapted to histamine, it was observed that in the animals with an active inflammatory process, there was an increase in the activity of this enzyme. This paper reports the results of an investigation into the alterations in the oxidative deamination of histamine during the course of inflammation.Because histamine may be oxidized by several plasma enzymes, e.g. ceruloplasmin [8], diamine oxidase [9], benzylamine oxidase [10], the term 'histaminase' is used in this paper to signify the oxidative deamination of histamine by plasma. Materials and methodsExperiments were performed on guinea-pigs of both sexes, weighing 300-600 g. The animals were fed on a standard pellet diet with addition of carrots.Inflammation: Two methods were used: (1) Subcutaneous injection of commercial turpentine oil in a dose of 0.5 ml per animal into the subscapular or abdominal area. To the control animals 0.5 ml of 0.85~ NaC1 were given. The bacteriological tests to prove the sterility of the inflammation consisted of culture of turpentine, blood and the inflamed tissue on both aerobic and anaerobic media.Only in cultures of two animals out of six, were bacterial infections found: in one of them -B. megaterium, and in the other -Staphyl. albus. These positive cultures were, according to the bacteriologists, probably the result of a contamination during the collection of the material.
