The Histaminase Activity of Ceruloplasmin

Abstract: Whether or not diamine oxidase (DAO) and histaminase are identical enzymes has been vigorously debated (1-6) and we have previously contributed to the controversy with the finding that DAO and histaminase values of plasmas from many primates do not correlate ( 7 ) . In further studies (8) we showed that acrylamide gel electrophoresis of plasmas from man and all of many primates examined separated an oxidase fraction which readily utilized histamine as a substrate, but not putrescine (as is characteristic of DA… Show more

“…In the total sample data set and in the SEM findings, the relationship of correlates between % free copper, zinc, and ceruloplasmin nicely fitted the known relationship that exists between excess free copper and low zinc and/or low ceruloplasmin levels (102). In an inflammatory setting, Cp is an acute phase reactant that can act as a diamine oxidase (ceruloplasminase) to enhance histamine metabolism and lower histamine levels (103). Elevated free copper, as found in this study, is also expected to support the activity of the copper containing enzyme diamine oxide (DAO), which also metabolizes histamine leading to lowered histamine levels (65, 104).…”

“…Thus o-dianisidine oxidation was the common denominator of both oxidative activities. As a consequence of the competition of both enzymes for the same indicator a histaminase activity was ascribed to ceruloplasmin at least [32]. Now the question arose, what enzyme was determined when the o-dianisidine test was used?…”

“…HAMPTON et al [32] reported a 'histaminase activity of ceruloplasmin'. Indeed, compared with blanks without histamine dihydrochloride an increased colour formation was observed when this substance was added to the incubation mixture.…”
