The skin is the largest organ in the body and plays several essential functions acting as a barrier that protects us from physical and chemical insults, prevents the entrance of pathogens and the loss of water, besides playing an esential role in the regulation of body temperature. The skin displays a high regenerative capacity, evidenced by its self-renewing activity and the process of wound healing, driven by the existence of several resident stem cell populations. Due to the high prevalence of skin pathology, and their anatomical accessibility, skin biopsies and their subsequent microscopic observation constitute a powerfull tool for the study of primary skin diseases, as well as cutaneous manifestations of systemic diseases. This gave rise to dermatopathology as a specific discipline that unifies dermatology and pathology. In this setting, staining with hematoxylin and eosin constitutes the gold standard method for microscopic observation and diagnosis. Yet, several additional stains are used for specific purposes, such as trichrome stains for the staining of collagens in the extracellular matrix. We have applied a recently developed stain (RGB trichrome, acronym for picrosirius Red, fast Green and alcian Blue) to human samples to assess the staining outcomes in normal skin tissues. RGB staining provides a high contrasted interface between epidermis and dermis, and a comprehensible staining of the different dermal structures such as blood vessels, nerves, and sweat and sebaceous glands. The specific staining of collagens by picrosirius red can be useful for the objective quantification of these proteins under polarized light microscopy. In hair follicles, RGB staining resulted in specific staining of the epidermal sheaths and the encasing connective tissue (i.e., dermal sheath). Interestingly, the dermal sheath show two domains in which blue predominates over red staining, thus indicating that proteoglycans prevail in these areas. These two zones are the dermal papilla and an uncharacterized zone located at the upper isthmus, that we have denominated as proteoglycan-enriched belt (PEB). While the role of the papilla in the induction and regulation of hair growth is clearly established, the possible role of PEB remains to be determined and merits additional investigation.

Hair loss, or alopecia, is a prevalent condition in modern society that imposes substantial mental and psychological burden on individuals. The types of hair loss, include androgenetic alopecia, alopecia areata, and telogen effluvium; of them, androgenetic alopecia is the most common condition. Traditional treatment modalities mainly involve medical options, such as minoxidil, finasteride and surgical interventions, such as hair transplantation. However, these treatments still have many limitations. Therefore, exploring the pathogenesis of hair loss, specifically focusing on the development and regeneration of hair follicles (HFs), and developing new strategies for promoting hair regrowth are essential. Some emerging therapies for hair loss have gained prominence; these therapies include low-level laser therapy, micro needling, fractional radio frequency, platelet-rich plasma, and stem cell therapy. The aforementioned therapeutic strategies appear promising for hair loss management. In this review, we investigated the mechanisms underlying HF development and regeneration. For this, we studied the structure, development, cycle, and cellular function of HFs. In addition, we analyzed the symptoms, types, and causes of hair loss as well as its current conventional treatments. Our study provides an overview of the most effective regenerative medicine-based therapies for hair loss.

Objective Human hair follicles (HFs) are populated by a rich and diverse microbiome, traditionally evaluated by methods that inadvertently sample the skin microbiome and/or miss microbiota located in deeper HF regions. Thereby, these methods capture the human HF microbiome in a skewed and incomplete manner. This pilot study aimed to use laser-capture microdissection of human scalp HFs, coupled with 16S rRNA gene sequencing to sample the HF microbiome and overcome these methodological limitations. Results HFs were laser-capture microdissected (LCM) into three anatomically distinct regions. All main known core HF bacterial colonisers, including Cutibacterium, Corynebacterium and Staphylococcus, were identified, in all three HF regions. Interestingly, region-specific variations in α-diversity and microbial abundance of the core microbiome genera and Reyranella were identified, suggestive of variations in microbiologically relevant microenvironment characteristics. This pilot study therefore shows that LCM-coupled with metagenomics is a powerful tool for analysing the microbiome of defined biological niches. Refining and complementing this method with broader metagenomic techniques will facilitate the mapping of dysbiotic events associated with HF diseases and targeted therapeutic interventions.
