The growth factor independence-1 transcription factor: New functions and new insights

Abstract: The growth factor independence-1 (Gfi1) transcription factor is required for proper development of neuroendocrine cells, sensory neurons, and blood. Patients with mutations in Gfi1 exhibit severe congenital neutropenia (SCN) or non-immune chronic idiopathic neutropenia of adults. Gfi1 was initially described as an oncoprotein that mediates tumor progression in a mouse model of leukemia; however, recent data suggest that Gfi1 may act as either an oncogene or an anti-proliferative tumor suppressor gene depending… Show more

“…This indicates that depending on the cellular context, Gfi1 functions as a tumor suppressor or oncogene. 82 Gfi1 can also cooperate with other oncogenes in lymphomagenesis. This is based on the fact that the time to tumor development on Gfi1 overexpression is significantly shortened when mice express other oncogenes like Myc and Pim.…”

“…Here it controls diverse developmental processes, such as cell fate determination, differentiation, proliferation and cell survival (for review, see (50)(51)(52)(53)(54)). Outside the hematopoietic system, Gfi1 mRNA has been detected in a wide range of other tissues, but expression of Gfi1 protein seems to be restricted to mechanoreceptors cells (hair cells and Merkel cells), neurons of the cerebellum and retina, and specialized epithelial cells in the gut and lung (Table 1).…”

“…G fi-1 encodes a nuclear zinc-finger transcription factor and was first identified as an integration site of Moloney murine leukemia virus in a screen for T cell interleukin-2-independent growth (1,2). Subsequently, Gfi-1 was found to be a frequent target of proviral insertion in T and B cell lymphomas.…”

“…Gfi-1 functions mainly as a repressor of transcription (1,2). It represses its target genes by binding to consensus DNA elements containing the AATC core sequence.…”
