The Glycoprotein α-Subunit is Critical for Secretion and Stability of the Human Thyrotropin β-Subunit

Abstract: TSH is a member of a family of heterodimeric glycoprotein hormones which have a common alpha-subunit but differ in their hormone-specific beta-subunit. To study the posttranslational processing and assembly of human TSH, eukaryotic expression vectors were constructed that contained either the human TSH beta gene only or both the TSH beta and alpha-genes. These vectors were transfected into Chinese hamster ovary cells and stable cell lines synthesizing TSH beta or TSH dimer were isolated. The kinetics of secret… Show more

“…Free TSH ␤-subunit, similarly to free LH ␤-subunit, is degraded intracellularly, and Ͻ10% produced is secreted into the culture medium. Therefore, simultaneous coexpression of ␣-subunit prevents intracellular degradation of TSH ␤-subunit (118). The most important domains in TSH expression and bioactivity recognized thus far are depicted in Figure 3.…”

“…These rare aberrant transcripts would not be expected to make functional protein because the folded tertiary structure of the a-subunit protein is crucial for binding to both [~-subunit and receptor (Lapthorn et al 1994). Secretion of the [3-subunits cannot occur without heterodimerization with the a-subunit (Corless et al 1987;Matzuk et al 1988Matzuk et al , 1989Keene et al 1989). Thus, the targeted disruption created a true null mutation for functional a-subunit in addition to the hormones TSH, LH, and FSH.…”
