O perfil antigênico de 45 herpesvírus (44 de bovinos, sendo seis amostras de referência de BHV-1 e 15 prováveis BHV-1; três amostras de referência de BHV-5 e 20 prováveis BHV-5) e uma amostra de herpesvírus bubalino (BuHV) foi examinado com um painel de anticorpos monoclonais (Acms) produzidos contra antígenos de herpesvírus bovinos. Para os exames, foi utilizada a prova de imunoperoxidase (IPX) sobre cultivos de células infectadas, tendo os Acms como anticorpos primários. A determinação dos padrões de reatividade das amostras de vírus frente aos Acms permitiu a diferenciação entre os tipos 1 e 5. Todas as amostras isoladas de casos de encefalite apresentaram perfil de BHV-5. Quatro amostras de BHV-5 isoladas de áreas geograficamente distintas apresentaram perfís de reatividade diferenciados em relação às demais amostras do tipo 5. Duas amostras de vírus com perfil antigênico de BHV-5 foram isoladas de sêmen de animais infectados. Estes resultados comprovam a utilidade da caracterização antigênica com este painel de Acms na tipagem de amostras de BHV-1 e BHV-5.

“…The BHV-5 isolate (named EVI 340/96) was tested with a panel of monoclonal antibodies capable of distinguishing BHV-1 from BHV-5 in immunoperoxidase monolayer assays as described (Silva et al 1996, Roehe et al 1997, Almeida et al 1998). Viral DNA was extracted from the virus isolate, as well as from control BHV-5 strains (V175 and N569; Souza et al 2002) as described by Van Oirschot et al (1995) and submitted to restriction endonuclease analysis (REA; Engels et al 1986). Restriction enzyme digestion with BstEII was performed under the conditions recommended by the manufacturer (BRL) and the DNA fragments separated by electrophoresis on a 0.5 per cent agarose gel.…”

-[Herpesvírus bovino tipo 5 (BHV-5) em bovino infectado pelo vírus da raiva. ] O encéfalo de um bezerro macho, de um ano de idade, que morreu com sinais de doença neurológica, foi submetido ao laboratório para diagnósti-co de raiva. O exame histopatológico revelou mielite moderada, meningoencefalite leve com infiltração perivascular e corpúsculos de Negri nas células de Purkinje do cerebelo. A infecção pelo vírus rábico foi ainda confirmada por imunofluorescência direta e por inoculação em camundongos. Além disso, um herpesvírus foi isolado dos tecidos do encéfalo. O vírus isolado foi antigênica e geneticamente caracterizado como Herpesvírus bovino tipo 5 (BHV-5). Não foi possível determinar se o BHV-5 teve algum papel ativo no desfecho da enfermidade, uma vez que o vírus poderia estar presente em forma latente nos tecidos neurais. Bovine herpesvirus 5 (BoHV-5) is a important cause of viral encephalitis in cattle in South America. Within the framework of developing a differential vaccine against BoHV-5, a deletion mutant was constructed based on a Brazilian BoHV-5 isolate. The target of the deletions were genes that code proteins implicated in the neurovirulence of BoHV-5, the glycoprotein I (gI), glycoprotein E (gE) and membrane protein US9. To construct the deletion mutant of BoHV-5, the flanking regions of all three genes were cloned in a prokaryotic plasmid. This deletion fragment was co-transfected with the viral DNA into bovine cells. Identification of deletion mutants was performed by immunostaining with an anti-gE monoclonal antibody. One of the gE negative viral populations found was purified, amplified and further examined by restriction endonuclesase analysis of its genomic DNA. The plaque sizes and penetration kinetics of the deletion mutant and wild type viruses were compared. The plaque sizes of the deletion mutant were significantly smaller than those of the parental strain (p ≤ 0.05), but no statistical differences were observed in penetration kinetics. The results indicate that the gI/ gE/US9 deletion mutant of BoHV-5 may have a reduced virulence in the host and is still viable enough to be a good candidate for the development of a BoHV-5 vaccine.
