The effects of fractionated megavoltage X‐irradiation on the rat submandibular gland: an assessment by electron microscopy

Abstract: Twenty-four rats were divided into groups that received cumulative doses of 500, 750, and lo00 rads of fractionated megavoltage X-irradiation. The animals sacrificed after three days showed acinar atrophy with isolated areas containing both severely and lethally damaged cells. The cells were undergoing apoptosis or showed the intracellular accumulation of vacuoles that coalesced and led to lysis. The pattern and severity of damage was dose-related. The animals sacrificed after 14 days had achieved a remarkable… Show more

“…In contrast to our findings, they found full recov ery of the gland within two weeks, with a return to preirradiation morphology. In accordance to our findings, there was no coagulative ne crosis or inflammatory response [20].…”

“…These findings support the results from Reade and Steidler [18] and Espinal et al [19], who found degranulation of the se rous cells within 2 h after irradiation and the process was completed after 2 days. Studies by Savage et al [20][21][22] describe a shrinkage of acinar mucous cells after 3 days. This result was not confirmed in our material, where there was only slight vacuolization in the mucous cells.…”

“…13,14 In the present study, IPR was administered to mice to release the secretory granules from submandibular gland acinar cells before irradiating them, so that the effect of irradiation on protein synthesis and secretion could be determined histochemically. Savage et al 15 exposed the submandibular glands of rats to a 10-Gy dose of X-ray, and noted marked morphological changes after 3 days, but not after 14 days. Kondow 16 irradiated the jaw (including the parotid gland) of mice with a 10-Gy X-ray dose and observed marked morphological changes in parotid gland organelles 3 days later.…”

“…Morphological studies of salivary glands following irradiation have used mainly conventional light and electron microscopical techniques (6)(7)(8)(9)(10). None of the studies, as far as we know, have dealt with the influence on the cytoskeleton, i.e.…”
