The effects of cyclooxygenase inhibitors on tumor-induced anorexia in rats

Abstract: Background. Interleukin‐1 (IL‐1) and tumor necrosis factor (TNF) are potent induces of prostaglandin (PG) synthesis and injection of PGE, IL‐1, or TNF decreases food intake in healthy animals, whereas the anorexigenic effects of injected IL‐1 and TNF are blocked by inhibitors of PG synthesis. It has been hypothesized that host secretion of IL‐1 and TNF contribute to tumor‐induced anorexia. This study was undertaken to determine whether administration of PG inhibitors alters food intake in anorectic rats implan… Show more

“…Walker 256 tumour has been considered to be a very well established model for the study of cachexia since the 1980s (Guaitani et al 1982). It induces most of the symptoms related to human cachexia and anorexia (McCarthy and Daun 1992), including alterations in the plasma concentration of most anabolic hormones (Seelaender et al 1996a) and marked changes in intermediary metabolism (Argilés and López-Soriano 1991;Seelaender et al 1996Seelaender et al , 1998.…”

“…The non-tumor control group received 1 ml of saline instead. Tumor weight in grams was estimated by an empirically-derived formula: length× width×1.33/100 (McCarthy and Daun, 1993). Tumor masses were excised at sacrifice, weighed and then processed for H/E staining to validate the tumor development in each animal (data not shown).…”

“…The Morris 7777 hepatoma was obtained from American Type Culture Collection (Line McA-RH7777; Bethesda, MD). A 1-mm tumor fragment was implanted between the scapulae with a trocar needle, as previously described (Fridriksdottir & McCarthy;McCarthy & Daun, 1993). The area between the scapulae was palpated daily for growth of the implanted tumor fragment.…”
