A BSTRACT In this study, acute toxicity of kenikir leaf ( Cosmos caudatus HBK) ethanolic extract was conducted on Wistar white male rats with fixed dose procedure. The extraction method used was maceration using 70% ethanol. A dose of 2000 mg/kgBW was determined as the starting dose based on the preliminary test result. The rats used in the main test were divided into the normal group and the 2000 mg/kgBW dose group, each used five animals in each group. The results of the test showed that there were no deaths or toxic symptoms both of the normal group and the 2000 mg/kgBW dose group. The conducted preliminary test results a dose of 2000 mg/kgBW as the starting dose. Next, main test is done toward two groups of rats: Normal group and 2000 mg/kgBW dose group each consist of five animals. The main test results in neither deaths nor toxic symptoms from those groups. The range of toxic doses of kenikir leaf ethanolic extract that can cause acute toxicity is >2000 mg/kgBW, and hence, this experiment classified in the practically nontoxic category. Statistical analysis on effect to macroscopic organs of the liver, heart, and kidney showed no significance ( P > 0.05) from kenikir leaf ethanolic extract. Average levels of biochemical parameters from the 2000 mg/kgBW group and the normal group were in the normal range. Ethanolic extract at a dose of 333 mg/kg BW does not cause severe pancreatic damage, and hence, it is considered safe for use as an antidiabetic.

Peroksidasi lipid akibat radikal bebas dapat menyebabkan kematian sel yang dapat merusak berbagai organ, termasuk liver. Hal ini dapat ditandai dengan peningkatan kadar malondialdehid. Pengukuran parameter ini pada darah dan organ digunakan sebagai penanda adanya stres oksidatif. Radikal bebas akibat peroksidasi lipid dapat diredam dengan senyawa antioksidan. Beberapa penelitian telah membuktikan aktivitas antioksidan tebu (Saccharum officinarum). Daun tebu dan jus tebu dari varietas yang berbeda-beda telah menunjukkan sifat antioksidan yang baik. Penelitian ini dilakukan untuk mengetahui efek ekstrak etanol tebu dengan berbagai dosis terhadap kadar malondialdehid plasma tikus yang diinduksi stress oksidatif dengan karbon tetraklorida (CCl4). Daun tebu diekstraksi dengan maserasi dengan pelarut etanol 96%. Pembuatan kurva baku malondialdehid dilakukan dengan seri larutan 1,1,2,2-tetraethoxypropane. Pengujian dilakukan terhadap hewan uji tikus Wistar yang dibagi menjadi beberapa kelompok. Pembagian kelompok meliputi kelompok normal tanpa induksi CCl4 diberikan CMC-Na 1%; kontrol negatif diberikan CMC-Na 1%; kontrol positif diberikan Silybum marinum yang 100 mg/kgBB; kelompok dosis yang terdiri atas 300, 400, dan 500 yang masing-masing diberikan ekstrak etanol daun tebu dosis dengan dosis 300 mg/kgBB, 400 mg/kgBB dan 500 mg/kgBB berturut-turut. Tikus diperlakukan sepanjang 14 hari lalu diambil sampel plasmanya pada hari ke-15 untuk pengukuran kadar malondialdehid. Hasil ekstraksi daun tebu mendapatkan rendemen sebesar 16,05%. Kurva baku yang digunakan untuk mengukur malondialdehid plasma tikus adalah y = 0,0059x + 0,0238. Hasil menunjukkan pemberian ekstrak daun tebu semua dosis memiliki nilai malondialdehid yang lebih rendah dibandingkan kontrol negatif. Dosis 400 mg/kgBB tidak memiliki perbedaan signifikan dengan kontrol positif, sedangkan dosis 500 mg/kgBB lebih rendah dibanding dengan kontrol positif. Hal ini diduga karena metabolit sekunder seperti flavon yang terdapat pada ekstrak yang dapat meredam radikal bebas. Ekstrak etanol daun tebu dapat berperan sebagai antioksidan dengan menurunkan kadar malondialdehid darah tikus terinduksi CCl4.

Objective: This study investigated the antihyperlipidemic and hepatoprotective effects of the hexane-ethanol fraction of methanol extract of Macaranga tanarius (HEM) in rats. Methods:The hexane-ethanol fraction was screened for toxicity by oral acute toxicity study. The antihyperlipidemic effect of the hexane-ethanol fraction and the unsolved of the hexane-ethanol fraction is measured against Wistar rats induced by glucose-fructose diets for 42 days through measuring serum cholesterol, triglyceride (TG), high-density lipoprotein cholesterol (HDL-c), low-density lipoprotein cholesterol (LDL-c), and fasting blood glucose. The hepatoprotective effect of the hexane-ethanol fraction is determined against Wistar rats with liver damage induced by carbon tetrachloride through measuring serum glutamate pyruvate transaminase (SGPT), serum glutamate oxaloacetate transaminase (SGOT), alkaline phosphatase (ALP), albumin, lactate dehydrogenase (LDH), and total bilirubin. Results:There is no toxic effect that was observed on acute toxicity study. The TG, LDL-c, and fasting blood glucose levels were significantly (p<0.05) reduced after both of treatment the hexane-ethanol fraction and the unsolved HEM. Administration of the hexane-ethanol fraction 68.6 mg/kgBW significantly (p<0.05) prevented elevation of SGPT, SGOT, LDH, ALP, and decreasing of albumin level. Conclusion:The study showed antihyperlipidemic and hepatoprotective activities of the HEM in animal models.
