The effect of total inorganic nitrogen and the balance between its ionic forms on adventitious bud formation and callus growth of ‘Węgierka Zwykła’ plum (Prunus domestica L.)

Abstract: The effect of different ammonium NH 4 + and nitrate NO 3 -ratios (4:1, 2:1, 1:1, 1:2, 1:4, 1:6) on organogenesis of 'Węgierka Zwykła' leaf explants cultivated on media with nitrogen levels equalling full-or half-MS was investigated. On media with total nitrogen equal to ½ MS, explant regeneration increased significantly and was highest on media with 1:2 or 1:4 NH 4 + :NO 3 -ratio. An excess of ammonium versus nitrate ions had a negative effect on both regeneration and biomass. Addition of potassium to the medi… Show more

“…The highest multiplication rate of A. polyphylla was achieved with intermediate concentrations of NH 4 ? and NO 3 -, namely ratios 20:40, 30:30 and 40:20, and decreased significantly outside these limits. Our results parallel those obtained in other studies (Avila et al 1998;Tsay 1998;Ramage and Williams 2002;Nowak et al 2007). Lowering the NH 4 ?…”

“…Conversely, in our experimental system, increasing the total N led to a proportional increase in the multiplication rate, which was not at the expense of shoot growth and quality. Similar results were reported for Prunus domestica (Nowak et al 2007).…”

“…All these alterations presumably enhanced and produced a balanced basal medium composition that was high suitable for callus formation. Obtaining a balanced medium composition was also necessary to successfully establish an in vitro culture of Gatharanthus roseus (Morard and Henry 1998), callus growth of Prunus domestica L. (Nowak et al 2007), regeneration of Dianthus henteri (Cristea et al 2010), and callus induction and regeneration of Foeniculum vulgare (Khorami and Safarnejad 2011). Specific basal medium for callus initiation in anther culture was also reported on other plants.…”

“…As a consequence of potassium uptake, nitrate use increases, promoting growth. Potassium nitrate was found to be important in the germination of plum (Prunus domestica L.) 27 while some banana has a preference for NH 4 NO 3 28 . MS medium has a high concentration of NH 4 NO 3 , which is a source of NH Nitrate-N induces the activity of nitrate reductase enzyme that reduces nitrate to ammonium, which is then incorporated into proteins 29,30 .…”

“…Additionally, the form of inorganic nitrogen and ammonium to nitrate ratio has been reported to greatly affect the growth and differentiation of cultured tissues 31,32 . Nevertheless, with a high NH + 4 , the uptake of cations is reduced and replaced instead by the uptake of an anion such as phosphorus with an accompanying release of an H + equivalent, which could lead to a more acidic pH and could be toxic to cells 27 . Glycine, present in MS medium, is also a source of organic nitrogen which has been reported to enhance plant regeneration in maize 33 .…”
