“…The Health Council of the Netherlands () set an Estimated Average Requirement (EAR) based on studies on vitamin B6 status assessed by plasma PLP concentrations, the tryptophan load test and α‐EAST in men (Harding et al., ; Baker et al., ; Yess et al., ; Miller and Linkswiler, ; Canham et al., ; Park and Linkswiler, ; Miller et al., ) and in women (Donald et al., ; Shin and Linkswiler, ; Brown et al., ; Driskell et al., ; Kretsch et al., ; Hansen et al., , , ; Huang et al., ). The Council considered a coefficient of variation (CV) of 20% for setting the Recommended Dietary Allowances (RDAs), due to uncertainty about the variation in the requirement (Hansen et al., ).…”

“…The reaction of the aldehyde group of pyridoxal (PL) or pyridoxal phosphate (PLP) with proteins during food processing has been shown to account for at least a portion of the losses of vitamin B6 reported in proteinaceous foods as a result of thermal processing or storage (Register et al 1950;Tomarelli et al 1955;Harding et al 1959). As previously discussed, interaction of PL or PLP with protein sulfhydryl groups has been reported to yield bis-4-pyridoxyl disulfide as a product Wendt and Bernhart 1960); however, the reductive bonding of these B6 compounds to amino groups which yields pyridoxyl amino compounds such as pyridoxyllsine appears to be a more quantitatively significant reaction Kirk 1977, 1978A;Gregory and Ink, unpublished).…”

“…This may be evi dence for great sensitivity of this test. On the other hand, the use of a relatively large dose of tryptophan, particularly if the diet already con tains a high level of protein, may represent an unrealistic demand on the vitamin B G activity of the tissues (84), and lead to an erroneously high estimate of requirements.…”
