The effect of platelet activating factor on different phases of murine in vitro fertilization

Abstract: The PAF concentration of 10(-6) M in sperm capacitation and sperm/oocyte coincubation yielded the greatest improvement in fertilization. However, continuing PAF treatment after sperm/oocyte coincubation had no beneficial effect on fertilization and preimplantation development.

“…This characteristic of albumin‐dependent release is apparently coupled with the spontaneous and reversible induction of sperm capacitation, because albumin is not generally recognized as a signaling molecule. In vitro capacitation induced by PAF has been further demonstrated in spermatozoa of the mouse model (Sengoku et al, 1992; Shi et al, 1992; Huo et al, 2000; Wu et al, 2001), the bovine model (Aravindakshan and Sharma, 1995), the stallion model (Odeh et al, 2003), and humans (Wu et al, 2001; Henkel and Schill, 2003).…”

“…The focus of the vast majority of these studies has been to observe and analyze the changes in spermatozoal motility and/or biochemical properties in the presence of exogenously added PAF. Upon incubation, effects of PAF on motility (Ricker et al, 1989;Hellstrom et al, 1991;Jarvi et al, 1993b), acrosome reactivity Angle et al, 1993;Fukuda et al, 1994;Krausz et al, 1994;Luconi et al, 1995), and fertilization potentials in vitro (Kuzan et al, 1990;Shi et al, 1992;Angle et al, 1993;Fukuda et al, 1994;Minhas, 1993;Sengoku et al, 1993) have been observed.…”

“…Although interesting, care must be used in drawing conclusions from such experiments. While some of these studies involve more ''physiological'' concentrations (low nM range) (Sengoku et al, 1993;Krausz et al, 1994), other effects were observed at higher concentrations of PAF (high nM to high µM range) Shi et al, 1992;Jarvi et al, 1993b;Minhas, 1993;Fukuda et al, 1994). The nanomolar range might even be an overestimation in semen since (Grigg et al, 1996) a Sequenced, either partially or completely.…”
