The effect of multimeric adiponectin isoforms and leptin on the function of rheumatoid fibroblast-like synoviocytes

Abstract: Adiponectin not only triggers pro-inflammatory and pro-destructive activities of rheumatoid FLS but also pre-disposes these cells to a stronger response to IL-1β. Thus, it is likely that adiponectin is more important in the initiation phase than in the chronic phase of RA.

“…Importantly, we revealed that HMW/MMW adiponectin triggers pro-inflammatory and pro-destructive activities of rheumatoid fibroblast-like synoviocytes (FLS) and also predisposes these cells to a stronger response to IL-1β. In addition, we found LMW adiponectin to incline rheumatoid FLS to a stronger response to IL-1β as well [28]. Altogether these findings point out pathogenic activity of adiponectin in RA that may explain its association with DAS28 observed in the present study.…”

“…Adiponectin exists in globular and multimeric (low molecular weight trimers [LMW], middle molecular weight hexamers [MMW], and high-molecular weight oligomers [HMW]) forms, that differ in their biological activities [26, 27]. We have previously reported that in RA sera the HMW isoform predominates, while in synovial fluid similar proportions of HMW and LMW are present [28]. Importantly, we revealed that HMW/MMW adiponectin triggers pro-inflammatory and pro-destructive activities of rheumatoid fibroblast-like synoviocytes (FLS) and also predisposes these cells to a stronger response to IL-1β.…”

“…However, LMW did not stimulate this secretion. This means that HMW both triggers inflammation and stimulates the release of IL-6 and IL-8, which is associated with pro-destructive effects in RA FLS [127]. The secretion of TGFβ, IL-6, IL-1Ra, PGE 2 , IL-8, and VEGF was considerably increased in HMW-treated RA-adipose mesenchymal stem cells but not in LMW-treated cells [128].…”
