INTRODUCTION:Recently, Nanosilver fluoride (NSF) has been raised in the research field of caries prevention. Pit-and-fissure sealants could have an additional antibacterial and remineralizing benefit through enamel pretreatment by NSF. OBJECTIVES:The aim of the study was to evaluate in-vitro the effect of NSF pretreatment on shear bond strength, marginal seal, and quantity of color change of pit and fissure sealant in comparison to sealant only in permanent teeth. MATERIALS AND METHODS: Fifty-four freshly extracted permanent teeth were collected and randomly allocated into two groups: Group I: (n=27) NSF was applied to the enamel prior to sealant application. Group II: (n= 27) enamel was etched and sealed. Each group was further divided into three subgroups according to the test applied. (n= 9 teeth each). Subgroup A: Shear bond strength was measured using a Universal Test Machine. The deboned area was then examined under a stereomicroscope to assess the mode of failure. Subgroup B: Microleakage samples were examined under a stereomicroscope to assess the degree of microleakage. Subgroup C: Color Assessment: After a sealant application, any change in sealant color was evaluated using a spectrophotometer. RESULTS: There was no statistically significant difference between both subgroups neither in shear bond strength nor microleakage. The comparison of the final color of sealants in both subgroups showed a higher color change in subgroup Ic with a significant difference (P-value =0.001). CONCLUSION: Pretreatment with nanosilver fluoride prior to pit-and-fissure sealant application did not affect neither its shear bond strength nor its sealing ability. However, it altered the sealant color to an extent that exceeded the clinical acceptance levels.

Aim: This study was designed to evaluate the antimicrobial efficacy of Nano Silver Fluoride, Fluoridated Self-assembling peptide, and Sodium Fluoride Varnishes on oral Streptococcus mutans bacteria. Materials and Methods:The Antimicrobial efficacy was evaluated using agar-disk diffusion method. Three Muller-Hinton agar plates cultured with Streptococcus mutans (ATCC1402) were prepared. Each plate received three remineralizing agents (Group 1: Nano Silver Fluoride, Group 2: Fluoridated Self-assembling peptide;and Group 3: Sodium Fluoride varnish; as a positive control group) on three filter paper discs. Then the plates were incubated anaerobically for 24 h at 37°C. The diameter of the formed inhibition zones was measured in millimeters by a ruler to assess the antimicrobial effect. Statistical analysis was done using Kruskal-Wallis test and Mann-Whitney post hoc Test. The level of significance was set at P ≤ 0.05.Results: Both Fluoridated Self-assembling peptide and Nano Silver Fluoride had antimicrobial effects against Streptococcus mutans. However, the median value of inhibition zone size (mm) was greater in Fluoridated Self-assembling peptide 21 (18-22) as compared to Nano Silver Fluoride 9 (8-9), with a statistically significant difference (P value = 0.046). Enamelast Sodium Fluoride varnish showed no inhibition zones.Conclusions: Fluoridated Self-assembling peptide had a better antimicrobial effect compared to Nano Silver Fluoride and conventional Sodium Fluoride varnish against Streptococcus mutans.

ÖZET Geçmişten günümüze birçok farklı meslekte ve sağlık sektöründe kullanılmakta olan gümüşün diş hekimliğindeki endikasyonları ve popülerliği giderek artmaktadır. Sahip olduğu antibakteriyel özelliği, karyojenik bakteriler üzerindeki bakteriostatik ve bakterisidal etkinliği çürük tedavisindeki kullanımını desteklemektedir. Florür ile bir arada kullanımı sinerjitik etki oluşturabilmektedir. İdeal özellikte bir ürün geliştirebilmek için farklı içeriklerde, kombinasyonlarda ve miktarlarda gümüş bulunduran materyallerle ilgili çok sayıda çalışma ve araştırma yapılmaktadır. Gümüş ve florür iyonlarını bir arada bulunduran gümüş florür ve gümüş diamin florür (GDF) içerikli ticari ürünler piyasada mevcuttur. Dünya Sağlık Örgütü, küçük çocuklarda oldukça sık görülebilen erken çocukluk çağı çürüklerinin tedavisinde GDF solüsyonunun kullanımını desteklediklerini belirterek, hem erişkinler hem de çocuklar için ilaç kategorisine aldıklarını açıklamışlardır. Ancak bu solüsyonların diş dokusunda siyah renklenmeye neden olabilmesi potasyum iyodür ve glutatyon gibi bileşiklerle birlikte kullanımını gündeme getirmiştir. The World Health Organization stated that they support the use of SDF solution in the treatment of early childhood caries, which can be seen quite frequently in younger children, and announced that they included it in the category of drugs for both adults and children.
