The dopamine receptor antagonist trifluoperazine prevents phenotype conversion and improves survival in mouse models of glioblastoma

Abstract: Glioblastoma (GBM) is the deadliest adult brain cancer, and all patients ultimately succumb to the disease. Radiation therapy (RT) provides survival benefit of 6 mo over surgery alone, but these results have not improved in decades. We report that radiation induces a glioma-initiating cell phenotype, and we have identified trifluoperazine (TFP) as a compound that interferes with this phenotype conversion. TFP causes loss of radiation-induced Nanog mRNA expression, and activation of GSK3 with consecutive posttr… Show more

“…Phenothiazine analogs have versatile applications across many diseases, including antimicrobial effects [ 24 , 25 , 26 ], antitumor effects [ 27 , 28 , 29 , 30 , 31 , 32 , 33 ], and potential roles in the treatment of neurodegenerative disease [ 34 , 35 , 36 , 37 , 38 , 39 , 40 , 41 ]. In the past, we showed PMZ binds to Abeta available in the brain lysate of 5XFAD mice in an in vitro study.…”

“…Accumulating evidence from various experimental models has implicated a molecular connection between the dysregulation of dopamine receptor biology and human cancers (32)(33)(34). In a recent study, we provided the first evidence demonstrating that DRD2 expression is inversely associated with clinical PCa progression.…”

“…This agent prevented the radiation-induced up-regulation of the Yamanaka factors, and decreased Sox2, and β-catenin protein levels. RNA-Seq analysis demonstrated that combination treatment affected cell cycle progression and cell division in GBM cells [16]. Second-generation antipsychotics, due to fewer side effects, also warrant further investigation for this devastating disease.…”
