The DNA Endonuclease Mus81 Regulates ZEB1 Expression and Serves as a Target of BET4 Inhibitors in Gastric Cancer

Abstract: DNA replication and repair proteins play an important role in cancer initiation and progression by affecting genomic instability. The DNA endonuclease Mus81 is a DNA structure-specific endonuclease, which has been implicated in DNA replication and repair. In this study, we found that Mus81 promotes gastric metastasis by controlling the transcription of ZEB1, a master regulator of the epithelialmesenchymal transition (EMT). Our results revealed that Mus81 is highly expressed in gastric cancer samples from patie… Show more

“…Multiple studies have also focused on BET inhibitors in cancer. In this context, it was observed that the DNA endonuclease Mus81, which regulates ZEB1, may be targeted by BET4 inhibitors (Yin et al, 2019). In general, miRNA-based therapeutic options targeting ZEB1 might represent promising tools for targeting ZEB1 but need to be further developed, and delivery methods and therapeutic agent stability should also be investigated with priority.…”

“…Genetic polymorphisms in MLH1 have also been associated with male infertility [178]. In addition, some of these DNA-remodeling enzymes, such as for example MUS81, are also emerging as potential targets in chemotherapeutic intervention [179][180][181][182]. Thus, advances in yeast studies have important implications in understanding the basis of human disorders linked to genome instability.…”

“…The IHC scores were calculated as described previously [26]. The staining intensity scores were as follows: 0 (negative), 1 (weak), 2 (medium), and 3 (strong); staining area scores were 0 (none), 1 (1-25%), 2 (26-50%), 3 (51-75%), and 4 (76-100%).…”

“…2c and d). As described previously, AGS cells showed low expression of MUS81 [26]. We further constructed the MUS81 wild-type (MUS81-WT) plasmid and transfected it into AGS cells (oeMUS81-WT), whereas the control group was transfected with the vector plasmid (oeVector).…”
