Fatty acid profiles of 202 coryneform and nocardioform bacteria were recorded by gas chromatography. Strains were grouped according to their profiles using mean linkage cluster analysis and similarity measures based on the correlation coefficient, the angular separation between vectors in a multidimensional space and the degree of overlap between superimposed traces. Comparisons using both real and hypothetical data showed the last of these measures to be the most effective. Strains were divided into two major groups, depending on whether they contained predominantly straight chain or iso-and anteiso-branched acids. The first group was divided into two subgroups according to the relative proportions of the characteristic acids present ; one subgroup had six clusters containing the rhodococci, nocardiae, mycobacteria and caseobacters, and the other had two containing the xanthobacters and true corynebacteria. The second group was divided into one subgroup containing strains of Arthrobacter simplex, Arthrobacter tumescens and Arthrobacter duodecadis, and one having three clusters. One cluster from this latter subgroup contained cellulomonads, one contained brevibacteria and curtobacteria and one contained arthrobacters, oerskoviae and kurthiae. Identification to generic level by fatty acid composition alone may not be feasible, but fatty acid analysis coupled with morphological examination may be sufficient to identify Corynebacterium, Arthrobacter, Cellulomonas, Oerskouia, Brevibacterium, Caseobacter, Kurthia and the A. simplexltumescens taxon. Distinction is not easy between Curtobacterium, Microbacterium and the diaminobutyric acid-containing coryneforms and between Rhodococcus, Mycobacterium and Nocardia.

Menaquinones were the only isoprenoid quinones found in 85 of the 95 coryneform bacteria examined. Dihydromenaquinones having nine isoprene units were the main components isolated from Corynebacterium bovis, from other glutamic acid-producing strains, and from Arthrobacter globiformis and related species. Dihydromenaquinones with eight isoprene units were found in Brevibacterium linens, the remaining Corynebacterium species and strains probably belonging to the genus Rhodococcus. Tetrahydromenaquinones with eight isoprene units were found in Arthrobacter simplex and Arthrobacter tumescens, and with nine isoprene units in Cellulomonas and Oerskovia. Kurthiu and Curtobacterium were characterized by menaquinones with seven and nine isoprene units, respectively, and Microbacterium lacticum and Corynebacterium aquaticum had comparable amounts of menaquinones with 10 and 11 isoprene units. Strains received as Brevibacterium leucinophagum, Corynebacterium autotrophicurn, Corynebacterium nephridii, Mycobacterium jlavurn, Mycoplana rubru and Protaminobucter ruber contained ubiquinones as their sole isoprenoid quinones. The isoprenoid quinone data correlate well with major trends in coryneform taxonomy and are of value in the classification of coryneform and related bacteria.

The classification of plant-pathogenic coryneform bacteria has been revised many times. All plant-pathogenic coryneform bacteria were once classified in the genus Corynebacterium. Currently, all are classified in the genera Arthrobacter, Cluuibacter, Curtobacterium or Rhodococcus. Difficulties still remain regarding the characterization of species and subspecies within these groups. Analysis of cellular fatty acids by high-resolution capillary gas chromatography has recently become a standard method of bacterial identification in many laboratories. In the studies reported here, cellular fatty acid analyses were performed to examine the plant-pathogenic coryneform bacteria as an autonomous group. A total of 189 bacterial strains were profiled. Results from these studies support the division of plant-pathogenic coryneforms into four major groups (genera). Further subdivision was not required for Rhodococcus and Arthrobacter, as each genus contains only one species of plant-pathogenic bacteria. Some, but not all, Curtobacterium pathovars could be differentiated from one another based on fatty acid analysis alone. Infrasubspecific classification of plant-pathogenic Curtobacterium organisms appears to be appropriate. Most of the species or subspecies belonging to the genus Cluuibacter could be differentiated using fatty acid profiles. Other chemotaxonomic methods also differentiate organisms within Cluuibacter. Reclassification is warranted within Cluuibacter michigunense. Cluuibacter michigunense subspecies should be returned to full species status.
