The Distribution and Common Amino Acid Polymorphisms of Human Papillomavirus (HPV)-31 Variants in 2700 Women from Northern China

Abstract: To investigate the distribution of Human papillomavirus (HPV)-31 A, B and C variants as well as the common amino acid polymorphisms in Chinese women, all 14 HPV-31 positive cervical exfoliated cell specimens identified from a descriptive study including ∼2700 women from Northern China were analyzed. HPV-31 positive specimens were identified by Mass Spectrometry and the fragments of partial Long Control Region, E6 and E7 were amplified and directly sequenced or cloned into vector and then sequenced to confirm t… Show more

“…However, this differs from the rate (0.4%) reported by a meta-analysis of females with normal cytology from Asia (28), which was in accordance with an international study that reported that the prevalence of HPV-31 is lower in Asia (0.3%) compared with the global average (0.8%), particularly in European (2.3%) and Latin American (1.2%) females with normal cytology (29). However, the rate of HPV-31 prevalence in Sichuan, China, differed from the 0.52% previously reported in Northern Chinese females (30). Future studies regarding geographic variation and ethnic differences may be worthwhile.…”

“…Chagas et al (35) examined five HPV-31 positive specimens from Brazil and reported that the proportions of A, B and C variants were 57.2, 5.7 and 37.1%, respectively (35). However, previous investigations have only observed variant lineages A (64.3%) and C (35.7%) in Southern China (30). As above, in the present study the variant lineage A of HPV-31 was the most commonly detected variant, followed by C. However, the present study also observed the presence of variant lineage B, which has not been previously reported from Southern China.…”

“…Both cases of HPV 16 belonged to HPV 16 lineage A, the case of HPV 31 belonged to HPV 31 lineage A, and the HPV 58 case belonged to HPV 58 lineage A. The lineage/sublineage classification of the 3 HPV types in HNC is consistent with the previous reports of HPV lineage distribution data in cervical cancer within China, 2330 indicating that the lineage/sublineage classification of the HNC causing HPV might be same with cervical cancer. Those data updated the lineage distribution data of HPV in HNC within China, which could be instrumental for designing HPV vaccines and HPV detection assays in China.…”

“…HPV E7 protein was divided into three main domains CR (conserved region)1, aa 1–15; CR2, aa 16–37; and CR3, aa 38–98, CR1 and CR2 mediate binding of viral oncoproteins to overlapping cellular proteins, including the product of retinoblastoma gene (pRB), cyclin A and cyclin E [ 32 , 33 ]. 21–29 associated with the binding of retinoblastoma (Rb) tumor suppressor, since E7 acts as the major immortalizing protein through Rb/E2F pathway, amino acid alternations in Rb binding domain may lead to the change of E7′s ability to immortalize cells among variants [ 34 ]. The CR3 domain contains two “Cys-X-X-Cys” sequences located at amino acid residues 58–61 and 91–94 participating in Zn binding, are the most important regions associated with the transforming ability of E7 protein [ 32 , 35 ].…”
