This work describes a simple photometric determination of the iodine concentration in thyroid tissue, a method based on the well-known catalytic Sandell-Kolthoff reaction. The modified ceric arsenite reaction is very sensitive and does not require complicated laboratory equipment. It can be performed in any routine clinical laboratory if instructions are followed strictly. By means of this method iodine concentration can be determined even in small specimens, for instance puncture samples of thyroid tissue or thyroid glands of small animals (mouse). Previous to the analysis, the tissue is digested in a mixture of sodium chlorate and perchloric acid at 100 degrees C. Using this manner of digestion between 94 and 110% of iodine in the sample were recovered. Comparison with neutron activation analysis showed excellent agreement of the obtained values.

The whole population of Certosa (Karthaus) (altitude 1327 m), a little village in the Alto Adige province in Northern Italy, was studied regarding the incidence and pathophysiological data of endemic goitre. The study included 204 subjects: in 85 % of the whole population, and in 48 % of the school-children population from 6-14 years of age, thyroid enlargement and/or nodularity was found. The 24 h [131I] uptake was 48.6 \m=+-\11.96; the grade "0" thyroids also showed an increased uptake. The region is poor in iodine; the mean iodine content of 55 samples of local drinking water was 0.81 \ m=+-\0.96 \g=m\g/I;the iodine content of several foodstuffs was definitely lower than those from Turin's markets. The mean iodine excretion in 60 samples of urine was 35.96 \ m=+-\ 22.4 \g=m\g/g creatinine. Urinary iodine excretion showed a linear negative correlation with [131I] uptake and did not correlate well with the presence or size of the goitre.The mean values of PBI (6.12 \ m=+-\ 1.57 \g=m\g/100ml) of T4 (7 \ m=+-\ 2.3 \g=m\g/100ml), of T3 (121 \ m=+-\ 55.4 ng/100 ml) and of a free thyroxine index (ETR = 0.95), as well as of TSH (2.63 \ m=+-\ 1.9 \g=m\U/ml) were in the normal range. Grade III goitres had slightly lower hormonal values, and a somewhat elevated T3/(T4 x 100) ratio (0.19). Serum TSH levels showed no correlation with the presence or size of the goitre, radioiodine uptake, the urinary iodine excretion, and not always showed an inverse correlation with the peripheral thyroid hormone values. A kinetic method for the determination of iodide based on its inhibitory effect on the Pd(III) catalysed reaction between ethylenediaminetetraacetic acid (EDTA)-Co(III) and the hypophosphite ion is described. The reaction was followed spectrophotometrically by measuring the decrease in the absorbance at 540 nm. Under the optimum experimental conditions of 2.6 x 10(-3) mol dm(-3)Co(III)-EDTA, 0.4 mol dm(-3)H2PO2-, pH 3.2 (adjusted with Britton-Robinson buffer), 0.57 micrograms ml(-1) Pd(III) and 20 +/- 0.2 degrees C, iodide was determined in the range 2-28 ng ml(-1). The method was applied to the determination of iodide in pharmaceutical products, iodinated salts, cow's milk and infants' powdered milk.
