The Cryopreservation of Striped Bass Morone saxatilis Semen

Abstract: Abstract.— Two experiments were designed to improve upon existing methods for cryopreserving striped bass Morone saxatilis, semen. In the first experiment, two extenders, two cryoprotectant concentrations, and two freezing rates were evaluated on the basis of post‐thaw semen motility after 1, 7, and 30 d of storage at −196 C. Semen samples cryopreserved at a freezing rate of −40 C/m resulted in a significantly higher percentage of motile sperm (P < 0.001) and longer duration of spermatozoa motility (P < 0.001)… Show more

“…Sodium chloride at concentration of 150 m m plus 5% DMSO proved to be a suitable diluent for spotted sea bass and striped bass sperm (Brown and Brown, 2000; Jenkin‐Keeran and Woods, 2002; Zilli et al, 2003, 2004, 2005). Billard (1984) and Sansone et al.…”

“…It is interesting to note that the best extender for the cryopreservation of striped bass milt was not the best extender for the short-term preservation of striped bass milt. When extenders 1, 2, and 5 were used to cryopreserve striped bass milt, extender 2 yielded significantly higher sperm motility and fertilization percentages than did extenders 1 and 5 (Jenkins-Keeran and Woods 2002).…”

“…Extender 1 has been used successfully in the cryopreservation of milt from rainbow trout Oncorhynchus mykiss and brown trout Salmo trutta (Stein and Bayrle 1978), as well as from Arctic char Salvelinus alpinus (Piironen 1993). Extender 2 is used by many researchers for the short-term, refrigerated storage and cryopreservation of striped bass milt (Brown and Brown 2000;Jenkins-Keeran et al 2001;Jenkins-Keeran and Woods 2002). Extender 3 was developed to mimic the mean osmolality, pH, sodium, and potassium measured from milt samples collected from 15 striped bass near the peak of the spawning season in 1997.…”

To ensure that germplasm from both species is protected, distributed and efficiently utilized, cryopreservation research being conducted on Morone species has delivered practical protocols for short‐term storage (Jenkins‐Keeran, Schreuders, Edwards & Woods 2001; Jenkins‐Keeran & Woods 2002a; He & Woods 2003a; He, Jenkins‐Keeran & Woods 2004) and long‐term cryopreservation (Jenkins‐Keeran & Woods 2002b; He & Woods 2003b, 2004a, b) of semen.…”
