The correlation between blood levels of ibuprofen and clinical analgesic response

Abstract: A clinical trial comparing ibuprofen, 400, 600, and 800 mg, with aluminum ibuprofen, 400 mg, and placebo was conducted in patients with moderate or severe pain subsequent to third molar extraction. Pain intensity ratings and ibuprofen serum levels were obtained at baseline, 30 minutes, 1 hour, and hourly thereafter for 3 hours. Pain intensity ratings were also obtained at hours 4, 5, and 6. Serum levels at 1, 2, and 3 hours correlated significantly with the log dose of ibuprofen (r = 0.35, 0.49, and 0.48, resp… Show more

“…In adults, a linear relationship between plasma ibuprofen concentration and the degree of pain relief was reported by Laska et al [15]. Since plasma concentrations of the active S isomer are low in infants relative to adults a higher dosage might be required in infants.…”

“…Dexibuprofen is rapidly absorbed, and peak serum concentration is achieved within 1.5-2 hours of oral administration [6]. It has been established there is a significant correlation between serum Dexibuprofen levels and the resultant degree of pain relief, particularly 1 hour after administration [7]. Figure 1) and Sodium benzoate NaC 6 H 5 CO 2 are the well-known preservatives that are used primarily for their bactericidal and fungicidal properties.…”

“…Thus, the present study aimed to close this gap. Among NSAIDs ibuprofen was chosen as a potent analgesic the doserelated analgesic effects of which have been studied intensively in a clinical setting [5,6]. The present investigation included the testing of placebo and two doses of rac-ibuprofen (400 and 800 mg).…”
