The classification and phylogenetic history of the Amaryllidaceae is reviewed since the dawn of molecular systematics in the 1990’s. The family is now recognized as comprising three subfamilies: Agapanthoideae, Allioideae, and Amaryllidoideae, of which the latter is the largest. The family likely had a Gondwanaland origin in what is now Africa. Agapanthoideae is monotypic, endemic to South Africa, and the first branch in the family tree of life; Allioidieae is sister to Amaryllidoideae. Four tribes are recognized in Allioideae: Allieae (monotypic, with nearly 1000 species of Allium across the Northern Hemisphere), Gilliesieae (5–7 genera in southern South America), Leucocoryneae (six genera mostly in southern South America), and Tulbaghieae (monotypic, with ca. 30 species endemic to South Africa). Amaryllidoideae is cosmopolitan, but mostly pantropical, consisting of 13 tribes. Centers of diversity occur in South Africa, South America and the Mediterranean region. The American clade is sister to the Eurasian clade (tribes Galantheae, Lycorideae, Narcisseae and Pancratieae) of the subfamily. The American Amaryllidoideae resolves as two monophyletic groups, 1) the hippeastroid clade (tribes Griffineae and Hippeastreae) and 2) the Andean tetraploid clade (tribes Clinantheae, Eucharideae, Eustephieae, and Hymenocallideae). Molecular analyses are reviewed for each main clade of the family, along with the resultant taxonomic changes. Directions for future studies are briefly discussed.

Nowadays, plant genetic resources are often at risk of loss and destruction. Geophytes are herbaceous or perennial species that are annually renewed by bulbs, rhizomes, tuberous roots, or tubers. They are often subject to overexploitation, which, combined with other biotic and abiotic stresses, can make these plants more vulnerable to a decline in their diffusion. As a result, multiple endeavors have been undertaken to establish better conservation strategies. Plant cryopreservation at ultra-low temperatures in liquid nitrogen (−196 °C) has proven to be an effective, long-term, low-cost, and suitable conservation method for many plant species. Over the last two decades, major advances in cryobiology studies have enabled successful explants of multiple genera and types, including pollen, shoot tips, dormant buds, and zygotic and somatic embryos. This review provides an update on recent advances and developments in cryopreservation and its application to medicinal and ornamental geophytes. In addition, the review includes a brief summary of factors limiting the success of bulbous germplasm conservation. The critical analysis underpinning this review will benefit biologists and cryobiologists in their further studies on the optimization of geophyte cryopreservation protocols and will support a more complete and wider application of knowledge in this area.

Amaryllidaceae alkaloid (AAs) biosynthesis has garnered significant attention in recent years, particularly with the commercialisation of galanthamine as a treatment for the symptoms of Alzheimer’s disease. A significant amount of research work over the last 8 decades has focused on the understanding of AA biosynthesis, starting from early radiolabelling studies to recent multi-omics analysis with modern biotechnological advancements. Those studies enabled the identification of hundreds of metabolites, the characterisation of biochemical pathway, an understanding of the environmental stimuli, and of the molecular regulation of these pharmaceutically and agriculturally important metabolites. Despite the numerous works there remain significant gaps in understanding their biosynthesis in Amaryllidaceae plants. As such, further research is needed to fully elucidate the metabolic pathway and facilitate their production. This review aims to provide a comprehensive overall summary of the current state of knowledge on AAs biosynthesis, from elicitation of transcription factors expression in the cell nucleus to alkaloid transport in the apoplast, and to highlight the challenges that need to be overcome for further advancement.
