Potato virus Y (PVY) the type member of the genus Potyvirus, occurs wodd-wide as isolates which differ in host range and the type of symptoms caused. The sequences of a 5' segment of viral RNA overlapping the 5' non-translated region (5'NTR) alone (ten isolates) or the 5'NTR and the adjacent P1 coding region (eight isolates) were established. These data were used to quantify the polymorphism in the Y-terminal part of the PVY genome. Nucleotide sequence identity between isolates ranged from 66-100 % in the 5'NTR and from 70-100% in the P1 coding region. The lowest amino acid sequence similarity between PVY P1 was 77%, illustrating the high variability of this protein in the PVY species. Phylogenetic trees based on either 5'NTR or P1

The complete nucleotide sequence of potato virus A (PVA) was obtained from six independent cDNA clones. The RNA genome of PVA is 9565 nucleotides long and contains one open reading frame (ORF) of 9177 bases encoding a large polyprotein of 3059 amino acids with a calculated M r of 340K. Seven potential proteinase NIa, one HC-pro and one P1 proteinase recognition sites were found in PVA polyprotein by searching for cleavage site consensus sequences amongst the potyvirus group. The non-coding region preceding the ORF is 161 nucleotides long. The termination codon is followed by a 227-nucleotide sequence. Overall nucleotide sequence identity compared with several completely sequenced potyvirus genomes is between 53 and 58 %, with overall amino acid sequence identity between 65 and 71%. When the putative amino acid sequences of individual proteins of PVA were compared with the corresponding proteins of other potyviruses, P1 and P3 appeared the least conserved (34 to 53 %) whereas the other proteins were in most cases from 63 to 80 % identical to each other.

Incidence and characterization of Potato virus V infections in Iran

A survey was carried out to determine the extent of Potato virus V (PVV) infection, together with other potyviruses, in Iran in both commercial and local potato varieties. We found a low incidence of PVV in commercial varieties compared to a local potato cultivar Zardi, in which we noted a PVV infection up to *32.9 %. We determined the genomic sequence 9,812 nucleotides of one isolate (KER.LAL.P) from cultivar Agria and the 3 0 -terminal sequence including coat protein (CP) gene of four additional isolates from cultivar Zardi. The Iranian isolate PVV KER.LAL.P was found to share 91 % sequence identity with the Scottish isolate DV-42 (AJ243766) of PVV. The CP gene sequences of the PVV isolates from Iran shared 96.5 to 99.3 % pairwise nucleotide identity and they shared \97.5 % pairwise identity with the CP sequences of all other PVV isolates available in public databases. Our host studies indicated that the Iranian PVV isolate had a narrower host range and infected Nicotiana debneyi and N. glutinosa test plants. Within the commercial varieties of potato in Iran, we noted a significant amount of mixed potyvirus infection. This study is the first report of occurrence and complete genome of PVV in Iran.
