The characterization of normal thyroid tissue by micro-FTIR spectroscopy

Abstract: In this paper, we report the spectral patterns of normal human thyroid tissue and methodology to interpret hyperspectral imaging data and protein conformational changes observed therein. Raw image datasets were imported into software written in-house in the MATLAB environment and processed to yield pseudo-color images of the tissue sections. All spectra were vector normalized, noise-filtered, and corrected for water-vapour contributions and scattering effects before being subjected to Hierarchical Cluster Anal… Show more

“…Development of a novel tool to discriminate between these fingerprints would be helpful to reveal hidden changes or uncover data that may help us to better understand modes of action. FTIR spectroscopy is a noninvasive technique that has been developed to detect the vibration of chemical molecules leading to various applications . Single‐cell FTIR is one of the applications which has been widely used for biological samples .…”

“…Unfortunately, the light source of Raman spectroscopy has been concerned in terms of poor sensitivity, weak light source and photodamaging. Nowadays, single‐cell FTIR has been developed to obtain chemical information from biological samples in terms of tissues and cells . Synchrotron light has been used as strong light source for increasing resolution of spectra.…”

“…A number of recent articles have covered using FT-IR imaging for tissue diagnostics in detail, with some preliminary work exploring thyroid tissue [911]. The technique holds promise as an adjunct tool to traditional histopathology for exploring the differences between FTCA and FVPCA.…”

“…Therefore it can be realized, for example, in the aforementioned Matlab environment. Matlab is one of the most popular environments designed for this type of analysis, both symbolic and numeric, in the areas of chemistry, bioengineering and computer science . Profiled, new scripts for preliminary hyperspectral image analysis together with the described source code, problems, solutions, and practical examples are proposed in this paper and discussed in the following sections.…”
