FTIR spectra signatures reveal different cellular effects of EGFR inhibitors on nonsmall cell lung cancer cells

Abstract: ATP‐analogue inhibitors, Gefitinib (Iressa) and Erlotinib (Tarceva) had been approved for advanced and metastatic nonsmall cell lung cancer (NSCLC) cells against tyrosine kinase domain of epidermal growth factor receptor (EGFR). Many techniques have been developed to better understand the drug mechanism which is multistep, time‐consuming and expensive. Herein, we performed Fourier‐transform infrared (FTIR) microscopy for evaluating the biochemical change on NSCLC (A549) cells after treatment. At levels that pr… Show more

“…Both microRaman and microFTIR spectra of A549 cells, prior to drug-administration, revealed a vibrational profile of the main cellular constituents coinciding with a healthy state ( Figure 4 ), according to the literature [ 15 , 42 , 43 , 44 ]. Cellular integrity was verified mainly by the presence of bands from nucleic acids (ν(OPO) backbone and ν s (PO 2 − ) characteristic from the native B-DNA conformation, at 786 and 1096 cm −1 , respectively), as well as from proteins (ν(C=O) Amide I/α-helix, at 1659 cm −1 ).…”

“…The FTIR device provides the spectra of compounds in the 400 cm -1 to 4000 cm -1 (2.5 to 25 μm) with a resolution of 2/cm. Jahangiri et al 77 , Piergies et al 78 , and Tabtimmai et al 79 measured the infrared spectrum of ERL at wavelengths 400 to 4000/cm. Figure 8 a illustrates the FTIR spectrum of the original ERL.…”

“…In recent years, studies of cellular behaviors after drug treatments in a single cell have received increased attention in anti-cancer research. Synchrotron-FTIR-MS provides a biochemical spectral signature for a specific drug treatment compared to a control group [ 12 , 16 , 17 , 34 ]. We then employed the Synchrotron-FTIR-MS analysis to present the effects of SG on biomolecular changes of CCA cells compared with the known drug, cetuximab, for potential use as an initial screening tool.…”
