The CD40-CD40L Dyad as Immunotherapeutic Target in Cardiovascular Disease

Abstract: Chronic inflammation drives the development of atherosclerosis. Despite optimal treatment of classical cardiovascular risk factors, a substantial portion of the population has elevated inflammatory biomarkers and develops atherosclerosis-related complications, indicating that a residual inflammatory risk drives atherosclerotic cardiovascular disease in these patients. Additional anti-inflammatory therapeutic strategies are therefore required. The co-stimulatory molecule CD40 and its ligand CD40L (CD154) have a… Show more

“…CD154, a marker of cardiac ischemia and the ligand of CD40, was often elevated in septic patients (McLean et al, 2008). Both CD154 and CD40 are potential therapeutic targets of cardiovascular disease (Bosmans et al, 2020). In our study, XBJ and C0127s significantly reduced the expression of CD40.…”

“…Another example is the CD40 gene. It is a member of the tumor necrosis factor receptor superfamily and encodes a protein playing a key role in a broad range of immune and inflammatory responses including T cell activation, immunoglobulin isotype switching and cytokine production [ 68 ]. CD40 has been implicated in various disorders, such as atherosclerosis, cardiovascular, metabolic (arterial hypertension, diabetes mellitus, etc.…”

“…CD40 has been implicated in various disorders, such as atherosclerosis, cardiovascular, metabolic (arterial hypertension, diabetes mellitus, etc. ), and the others [ 68 , 69 ]. Importantly, the CD40 DNA methylation levels [ 70 ] and CD40 expression (protein and mRNA) [ 71 ] were associated with gastric cancer.…”

“…It enhances leukocyte recruitment to the sites of vascular inflammation. In advanced lesions, it stimulates macrophages to increase production of matrix metalloproteinases (MMP) [ 7 ], particularly MMP-9, which is responsible for plaque destabilization and rupture [ 8 ]. Monocyte chemoattractant protein -1 (MCP-1) is another protein that promotes plaque vulnerability.…”
