ABSTRACT. With high nutritional value in its fruits, Dangshan Su pear has been widely cultivated in China. The stone cell content in fruits is a key factor affecting fruit quality in pear, and the formation of stone cells has been associated with lignin biosynthesis. O-Methyltransferase (OMT) is a key enzyme involved in lignin metabolism within the phenylpropanoid pathway. Here, we screened 26 OMT genes from the Pyrus bretschneideri cv. Dangshan Su genome using the DNATOOLs software. To characterize the OMT gene family in pear, gene structure, chromosomal localization, and conserved motifs of PbOMTs were analyzed. PbOMTs were divided into two categories, type I (designated PbCCOMTs) and type II (designated PbCOMTs), indicating the differentiation of function during evolution. Based on the analysis of multiple sequence alignment, cis-element prediction, and phylogenetic relationships, two candidate genes, PbCCOMT1 and PbCCOMT3, were selected for the analysis of temporal and spatial gene expression in pear. The promoter regions of both PbCCOMT1 and PbCCOMT3 contain regulatory motifs for lignin synthesis. Moreover, the two genes show high similarity and close phylogenetic relationships with CCOMTs in other species. Expression analysis showed that transcript levels of two PbCCOMTs were positively associated with the contents of both stone cells and lignin during the development of pear fruit. These results suggest that PbCCOMT1 and PbCCOMT3 are closely associated with lignin biosynthesis. These findings will help clarify the function of PbOMTs in lignin metabolism and to elucidate the mechanisms underlying stone cell formation in pear.

Summary Fruit is seed‐bearing structures specific to angiosperm that form from the gynoecium after flowering. Fruit size is an important fitness character for plant evolution and an agronomical trait for crop domestication/improvement. Despite the functional and economic importance of fruit size, the underlying genes and mechanisms are poorly understood, especially for dry fruit types. Improving our understanding of the genomic basis for fruit size opens the potential to apply gene‐editing technology such as CRISPR/Cas to modulate fruit size in a range of species. This review examines the genes involved in the regulation of fruit size and identifies their genetic/signalling pathways, including the phytohormones, transcription and elongation factors, ubiquitin‐proteasome and microRNA pathways, G‐protein and receptor kinases signalling, arabinogalactan and RNA‐binding proteins. Interestingly, different plant taxa have conserved functions for various fruit size regulators, suggesting that common genome edits across species may have similar outcomes. Many fruit size regulators identified to date are pleiotropic and affect other organs such as seeds, flowers and leaves, indicating a coordinated regulation. The relationships between fruit size and fruit number/seed number per fruit/seed size, as well as future research questions, are also discussed.

Sesame Fusarium wilt (FW) is a worldwide and hardly controlled fungus disease in sesame. Till now, the molecular resistance mechanism of sesame to FW remains unknown. The two resistant and susceptible sesame varieties, that is ‘Yuzhi 11’ and Rongxian black sesame (RXBS), were inoculated with Fusarium oxysporum f. sp. sesami (Fos) pathogens for 0, 6, 24 or 24 and 48h, respectively. Then, transcriptome samples were sequenced and surveyed systematically using RNA‐seq technology. The Kyoto Encyclopedia of Genes and Genomes (KEGG) analysis showed that ‘phenylpropanoid biosynthesis’ pathway may play more important role in sesame resistance after Fos inoculation. Furthermore, qRT‐PCR results demonstrated that the phenylpropanoid pathway with an emphasis on the synthesis of lignin contributes to the sesame disease resistance. Moreover, activity of phenylalanine ammonia lyase (PAL) and peroxidase (POD) proved the differences of gene expression patterns in the two sesame phenotypes. Overall, we speculated that the difference of FW symptoms between the resistant and susceptible plants might depend on whether the plants could activate the pathway timely and efficiently or not. The findings would help to clarify the molecular mechanism of sesame resistance to FW.
