The bone mass density in postmenopausal women using hormonal replacement therapy in relation to polymorphism in vitamin D receptor and estrogen receptor genes

Abstract: (1) There is no relationship between VDR and ERalpha genes polymorphism and the stage of osteoporosis related to the spinal BMD value before treatment. (2) The XX, PP or Bb markers or only X, P, B alleles are connected with a significant decrease of treatment efficacy. (3) Estradiol serum concentration before and during HRT is not dependent on the polymorphism of VDR and ERalpha genes.

“…The effectiveness of EP therapy is mostly determined by the ability to respond to hormonal substitution. This was confirmed both in studies 17 19 and in experiments by other authors 20 – 22 . All suggesting the involvement of the ER-α gene polymorphisms in determining response to oestroprogestagen therapy.…”

“…Studies on postmenopausal women subjected to hormonal therapy did not reveal which genetic variants of ER-α were predictors of poor BMD response to treatment. Depending on the study, either the xxPP haplotype [24, 25] or the XX genotype of XbaI [26, 27] and PP genotype of PvuII [27] were revealed as negative predictive factors. Other authors, in turn, did not observe significant relationships between ER-α and the response to hormonal treatment [2830].…”

“…[8][9][10]22 In another study, subjects with the PP and XX genotypes reveal decreased treatment efficiency. 5 To our knowledge, only one study has examined the relationship between ER polymorphisms and bone effects of phytoestrogen and found that low-dose isoflavone (30 mg d −1 ) supplementation is useful for maintaining BMD in premenopausal women with the XX genotype. 23 Our findings indicated that the association between the dietary phytoestrogen intake and BMD in postmenopausal women varied with ER polymorphisms.…”

“…1,2 Phytoestrogen is a natural estrogen-like substance that is gaining increasing attention as a possible alternative to estrogen. 5,6 As the hub of estrogen action, the ER gene is an important candidate gene for exploring the etiological and therapeutic diversities of PMOP. Given the similar chemical structure of phytoestrogen to that of endogenous estrogen, it can bind to estrogen receptors (ERs) and produce varying estrogenic effects.…”
