Infertility is a worldwide problem affecting couples that have unprotected intercourse, so the current study investigated the possible ameliorative effect of intra testicular stem cell injection in rats subjected to gentamicin induced testicular degeneration. For this study 100 male rats were randomly allocated into 4 groups, namely, the control group (group I), gentamicin group (group II), stem cell group (group III) and gentamicin and stem cell group (group IV). Gentamicin was administered daily for 2 weeks (80 mg/kg) to groups (II and IV). Stem cells were injected intra testicular into rete testis once by the end of gentamicin administration in group (IV) or after I/P injection of saline in group III. Blood and testicular samples were collected by the end of weeks 2, 6 and 10 from group (I) and (II), and by the end of weeks 6 and 10 from group (III and IV). The obtained results showed a significant decreases in serum testosterone, testicular testosterone and estradiol, sperm counts, sperm viability and an increase in % of fragmented DNA (indicating degenerative and necrotic changes) in testicular tissue of group (II) compared to group I , III and IV. Mesenchymal stem cells administration significantly improved all altered testicular parameters, where values of group IV (except testicular estradiol) were not significantly different from those of control group (I). Conclusively, mesenchymal stem cells proved to be beneficial in rats subjected to gentamicin induced testicular dysfunctions.

Abstract. The present study aimed to investigate the effects of melatonin (MT) on liver function and lipid peroxidation following hepatic ischemia-reperfusion injury (IRI). A total of 66 male Sprague-Dawley rats were randomly assigned into three groups: Normal control (N) group, ischemia-reperfusion (IR) group and the MT-treated group. A hepatic IRI model was developed by blocking the first porta hepatis, and subsequently restoring hepatic blood inflow after 35 min. Following reperfusion, changes in the levels of alanine aminotransferase (ALT), aspartate aminotransferase (AST), lactate dehydrogenase (LDH), malondialdehyde (MDA), superoxide dismutase (SOD) and glutathione (GSH) were detected by a chemical method at various time points. In the MT group, the MDA levels were significantly reduced (P<0.05) at all time points, as compared with the IR group. Furthermore, SOD activity was significantly increased (P<0.05) in the MT group, as compared with the IR group at all time points; and the levels of GSH in the MT group were significantly higher (P<0.05) than those of the IR group at 2, 4, and 8 h post-reperfusion. The levels of ALT, AST and LDH were significantly reduced in the MT group at each time point, as compared with that of the IR group (P<0.05). In conclusion, MT exhibits potent antioxidant properties that may create favorable conditions for the recovery of liver function following IRI. IntroductionIn order to effectively control bleeding during hepatobiliary surgery, hepatic portal occlusion is often used, as it can temporarily interrupt the portal vein and hepatic artery inflow. This method is widely accepted since it is clinically simple, practical and effective; however, there is an inherent risk of subsequent liver ischemia-reperfusion injury (IRI). Therefore, in recent years, clinical research in the field of hepatic surgery has focused on how to reduce blood loss and improve the safety of hepatic portal occlusion, whilst simultaneously reducing the risk of liver IRI.Previous studies investigating tissues and organs such as the brain, heart, kidney and retina have demonstrated that numerous free radicals are produced during IRI, when the endogenous radical scavengers cannot function appropriately, leading to cell damage (1,2). Therefore, investigating effective drugs that alleviate IRI-induced functional liver damage, and improve the postoperative survival rate, has become a research hotspot in the field of hepatic surgery. Melatonin (MT) is a neural endocrine hormone that is secreted by the pineal gland (PG), which exerts known antioxidant, antitoxic, anti-stress and anti-inflammatory effects (3,4). Testicular torsion (TT) is a medical emergency that primary affects newborns and young adolescents. It causes testicular injury due to the torsion of the spermatic cord and its components, initially in the venous blood flow and finally in the arterial blood flow. Prompt diagnosis and early surgical management are necessary in managing this urgent situation. The process of the pathophysiological events in ischemia-reperfusion is multifactorial and deals with the perception of the oxidative stress responsible for the consequences of ischemia/reperfusion (I/R) stress following TT. Duration and severity of torsion also play a significant role in the oxidative stress. A detrimental result of the defense system of the testes takes place resulting finally in testicular atrophy and impaired function. Antioxidant factors have been experimentally studied in an effort to front this state. They have been classified as endogenous or exogenous antioxidants. Endogenous antioxidants comprise a structure of enzymic enzymatic and non-enzymic enzymatic particles presented within cytoplasm and numerous other subunits in the cells. Exogenous antioxidants include a variety of natural and pharmaceutical agents that may prevent or ameliorate the harmful effects of I/R injury. In this study we review those factors and their ability to enhance the oxidative status of the testis. A feature insight into where we are heading is attempted.
