The autoregulatory translational control element of poly(A)-binding protein mRNA forms a heteromeric ribonucleoprotein complex

Abstract: Repression of poly(A)-binding protein (PABP) mRNA translation involves the binding of PABP to the adenine-rich autoregulatory sequence (ARS) in the 5′-untranslated region of its own mRNA. In this report, we show that the ARS forms a complex in vitro with PABP, and two additional polypeptides of 63 and 105 kDa. The 63 and 105 kDa polypeptides were identified, as IMP1, an ortholog of chicken zip-code binding polypeptide, and UNR, a PABP binding polypeptide, respectively, by mass spectrometry of the ARS RNA affin… Show more

“…All qRT-PCR primers for the UPL system were designed using the Roche Applied Science online assay design centre. The primers used for the SYBR Green system were as follows: E-cadherin (Tang et al, 1994), Snail sense (5Ј-CCTCAAGATGCACATCCG-AAGCCA-3Ј) and antisense (5Ј-AG-GAGAAGGGCTTCTCGCCAGTGT-3Ј), and ␤-actin (Patel et al, 2005). All primers were purchased from Invitrogen (Burlington, ON, Canada).…”

“…We hypothesized that the mechanism by which R-motif affects translation is likely through association with poly(A)-binding proteins (PABs) because these proteins have been shown to bind to not only poly(A) tails of transcripts to enhance translation, but also A-rich sequences located in their own 5′ leader sequences to inhibit translation 17 . To test our hypothesis, we examined the role of class II PABs (i.e., PAB2, PAB4 and PAB8) 18 by co-expressing PAB2 with three R-motif-dependent genes, ZIK3, BET10 , and SK2 and one R-motif-independent gene, SAC2 .…”
