The association between RCAS1 expression in laryngeal and pharyngeal cancer and its healthy stroma with cancer relapse

Abstract: Background: The purpose of this study has been to establish the level of RCAS1 -a membrane protein expressed in various cancer cells and able to induce apoptosis of CTLs and NK cells in pharyngeal and laryngeal cancer and its clear surgical margin -with respect to clinicopathological features and to patient's follow up and evaluate its possible role in cancer relapse.

“…This incidence of RCAS1 positivity is among the highest reported incidences for all types of head and neck malignancies, including oral and esophageal SCC [1825]. A similar incidence of RCAS1 positivity was reported in laryngeal and pharyngeal SCC, in which RCAS1 protein was detected in all the examined tissue samples [33]. To this respect, current substantial evidence supports that RCAS1 may be considered as a promising target for future (gene) therapeutic approaches [6,17].…”

“…Interestingly, the results of all recently published studies are very controversial. As yet, overexpression of RCAS-1 in HNSCC has only been demonstrated in 4 independent studies in oral squamous cell carcinomas [18][19][20][21] . Toyoshima et al [20] could not demonstrate a difference in survival between patients with low and high expression of RCAS-1 whereas Tsai et al [18] and Fukuda et al [19] showed a large survival benefit for patients with low expression of RCAS-1.…”

Dutsch-Wicherek et al [21] examined clear surgical margins in HNSCC patients and found significantly higher levels in patients with recurrences.…”

“…Although several researchers have suggested a potential link between advanced malignancy and high-level expression of EBAG9 in tumor tissue (3,(23)(24)(25)(26)(27)(28), evidence that this molecule itself exerts an in vivo tumor-promoting function has been lacking. Recently, Ogushi et al reported for the first time that the enhanced expression of EBAG9 in Renca mouse renal cancer cells leads to accelerated in vivo cell growth, using two representative clones generated by transfection of a human EBAG9 cDNA (12).…”
